| Objective:This study was aimed to examine the tumor suppressing role of LNC00472 in human HCC cells and clarify the mechanism of LNC00472 suppresses hepatocellular carcinoma cell proliferation,migration and invasion through miR-93-5p/PDCD4 pathway.Methods:1.Expression levels of LNC00472 in human HCC tissues and cells were detected using RT-qPCR.2.MTT assay,cell count assay,cell migration,cell invasion assay and cell colony formation assay were used in this study to examine cell proliferation and metastasis of human HCC cells.3.Luciferase assay,stably transfection and western blot were used to clarify the relationships between miR-93-5p and LNC00472,PDCD4 and miR-93-5p,LNC00472 and PDCD4,respectively.4.For RT-qPCR,cell functional experiments,flow cytometry and luciferase reporter assay,two-tailed t test was used in statistical analyses.For patient overall survival analyses,Kaplan-Meier curves and log-rank test were used.Results:1.LNC00472 is down-expressed in hepatocellular carcinoma cell and decreased LNC00472 expression predicts poor prognosis in HCC patients.2.Forced expression of LNC00472 inhibits hepatocellular carcinoma cellproliferation,migration and invasion.3.MiR-93-5p was a direct target of LNC00472,and miR-93-5p directly targeted PDCD4;LNC00472 suppressed proliferation,migration and invasion of human HCC cells through PDCD4.Conclusion:In summary,the present study systematically examined the tumor suppressing role of LNC00472 in human HCC cells.Low expression level of LNC00472 of HCC patients was correlated with poor prognosis.LNC00472 suppressed proliferation,migration and invasion of HCC cells.MiR-93-5p was a direct target of LNC00472,and miR-93-5p directly targeted PDCD4.ThemiR-93-5p/PDCD4 pathway mediated the suppressing role of LNC00472 in HCC cells.As an important tumor suppressor,LNC00472 could be used as a bio-marker for HCC therapy. |
