Blaps rynehopetera Fairmaire is a common folk medicine insect,which has long been used as a traditional medicineof YI nationality.It has been used to treat inflammation,infection and cancer.In addition,in the folk medicine prescription,there is a method of treating tumors by consuming living B.rynehopetera.On a global scale,malignant tumors are a very serious public health problem.Both the literature reports and our previous studies have demonstrated that B.rynehopetera has anti-tumor activity in cancer cells including AGS(Human gastric cancer cell),Coca-2(Human colon adenocarcinoma),HepG2(Human hepatoma cell)and U251(Human glioma cell),but there isn't any report about anti-tumor compounds from B.rynehopetera.In this experiment,the living B.rynehopetera was executed by 95%alcohol and the combined extracts were concentrated.It extracted with petroleum ether,ethyl acetate and n-butanol respectively,to obtain three extracts.The three extracts were assayed by SKBR3(Human breast adenocarcinoma),MD-A-MB-231(Human breast cancer cell),ASPC1(Human pancreatic adenocarcinoma)and PC3(Human prostate cancer cell).Results showed that ethyl acetate fraction had activity.It had antitumor activity at a concentration of 5 ?g/ml.14 compounds were isolated and purified from the ethyl acetate extract phase with antitumor activity.The structures of 9 compounds were identified by spectroscopic analysis:hydroquinone(1),2-methyl-1,4-benzenediol(2),2-ethyl-1,4-benzenediol(3),17-methyl-5-octadecenoic acid(5),(E)-(E)-undec-4-en-l-yl tridec-5-enoate(6),cholesterol(7),2-(2-hydroxyethyl)benzene-1,4-diol(9),1-(3,4-dihydroxyphenyl)isochroman-6,7-diol(10)and 2,5-dihydroxyphenethyl 2-hydroxypropanoate(11).Among them,compound 11 was isolated for the first time from B.rynehopetera.Compounds 1,2,3,9,10,and 11 have inhibitory activity against tumor cell MD-A-MB-231.And IC50 values of compound 1,2 and 3 anti-tumor cell MD-A-MB-231 activity were:69.86 ?g/mL,6.76?g/mL,2.6 ?g/mL;the inhinitory concentrations of compund 9,10 and 11 antu-tumor cell MD-A-MB-231 were IC50>40 ?M.The compounds having antitumor activity isolated from B.rynehopetera are all phenol derivatives.By measuring the antibacterial activity of the isolated compound,the results show that compounds 1,2,3 and 9 had antibacterial activity against P.aeruginosa.and the other compounds had no antibacterial activity.
