Objective In order to explore the clinical significance of the serum interferon-?3 level in chronic hepatitis B infection and its association with nucleos(t)ide analogue(NUC)administration,we measured interferon-?3 levels in sufferers with chronic hepatitis B.Methods The expression levels of interferon-?3 in serum of 119 sufferers with chronic hepatitis B and 22 healthy subjects were measured by ELISA.Detection of relative expression of IFN-?3 m RNA in peripheral blood mononuclear cells of patients with chronic hepatitis B by RT-PCR.The relationship between serum IFN-?3 level and serum serological markers and liver enzymes in patients with chronic hepatitis B.The relationship between serum interferon-?3 level and clinical manifestations in sufferers with chronic hepatitis B was analyzed.Peripheral blood mononuclear cells from healthy blood donors were cultured with nucleos(t)ide analogues for 48 hours,and cell culture supernatants were collected to investigate the effect of nucleos(t)ide analogues on the expression of interferon-?3 protein in peripheral blood mononuclear cells in vitro.Results Serum levels of IFN-?3 in sufferers with chronic hepatitis B were much lower than those in healthy people[14.7(5.9 ~28.5)pg/ml vs 76.1(39.4 ~112.0)pg/ml,Z =-10.114,P < 0.001 ].And the relative expression of IFN-?3 m RNA in peripheral blood mononuclear cells of patients with chronic hepatitis B was lower than that of normal people(Z=-2.25,P=0.0234).There was no significant correlation between serum IFN-?3 levels and ALT in AST(r=0.0089,P=0.5946),AST(r=0.03216,P=0.3120).Serum IFN-?3 levels in untreated chronic hepatitis B sufferers were associated with log HBV-DNA(r=-0.621,P <0.01),no significant correlation with HBs Ag r=-0.136,P=0.44,(P>0.05);HBe Ag positive chronic hepatitis B sufferers with serum IFN-?3 levels were lower than HBe Ag negative sufferers,the difference was statistically significant [16.4(9.0,31.3)pg/ml vs 8.7(2.9,23.2)pg/ml,Z=-2.205,P=0.02)];Serum IFN-?3 levels were significantly higher in patients treated with adefovir dipivoxil antiviral therapy than the patients treated with entecavir,tenofovir,telbivudine and lamivudine(P<0.05).The supernatant of peripheral blood mononuclear cells treated in vitro did not show higher IFN-?3 level.Conclusion The serum IFN-?3 level of sufferers with chronic hepatitis B is significantly lower than that of healthy people.The anti-viral treatment of adefovir dipivoxil has higher serum IFN-?3 level.
