Objective To investigate whether the protective effect of exogenous RBM3 on acute brain injury in rats,and whether the protective effect is similar to that of mild hypothermia on acute brain injury.Methods Forty Sprague-Dawley(SD)rats were randomly divided into four groups,10 rats a group.They are the normal control group(NC group),the brain injury group(CI group),the brain injury +RBM3 group(CR group),and the brain injury +mild hypothermia group(CM group).Groups(CI group,CR group,CM group)all constructed the acute brain injury model in oxygen method,the CM group kept rats anal temperature 32 ℃in mild hypothermia method.The neurobehavioral manifestations of rats in each group were observed and compared after 24 hours.The liver and kidney functions(CK,AST,Cr)and blood gas levels of the rats in each group were measured after 24 hours after the experimental treatment.The pathological damage of brain,lung,liver and kidney was observed by HE staining.The data were analyzed by statistical software.Results1.Acute brain injury model of rats was successfully constructed by hypoxia method.During it,rats showed cyanosis,restlessness,coma,death and other manifestations of hypoxia,neurological dysfunction and severe body injury.After 24 hours of hypoxic stimulation,compared with the normal control group,rats in the brain injury model group showed behavioral changes and decreased abilities in learning,memory and spatial orientation.2.Blood samples were collected for blood gas analysis(PH,PO2,PCO2,SaO2):(1)There was no statistical significance of difference in PH value between the groups.(2)There was statistical significance of the results of PO2,PCO2 and SaO2 in the brain injury group compared with those in the normal control group.(3)PO2 and SaO2 results were compared with those of the brain injury group after RBM3 or mild hypothermia treatment,and P<0.05 was found between the two groups,indicating a significant difference.(4)The results of PCO2:In RBM3 group and brain injury group showed no significant difference(P>0.05).The result of mild hypothermia group was significantly lower than that of brain injury group(P<0.05),and the difference was significant.3.Blood samples were collected for biochemical indexes(CK,AST,Cr)analysis.In that,CK,AST and Cr in the brain injury group were significantly higher than those in the normal control group,and after RBM3 or mild hypothermia treatment:(1)The CK results were lower compared with those of the brain injury group(P<0.05),and the difference between the two groups was insignificant(P>0.05).(2)The AST results were also lower compared with those of the brain injury group(P<0.05),and the difference between the two groups was significant(P<0.05).(3)Compared with the brain injury group,the results of Cr values in the RBM3 group were insignificant(P<0.05),and the results in the mild hypothermia group were significantly reduced(P<0.05).4.Comparison of the protective effects of important organs: HE staining results showed that the pathological images of brain,liver,lung and kidney of rats in the hypoxictreated brain injury group were significant damaged,while the degree of physical damage of brain,liver and lung was significantly reduced after RBM3 or mild hypothermia treatment.Conclusion1.Exogenous RBM3 has a protective effect on rats with acute brain injury.2.The protective effect of exogenous RBM3 on rats with acute brain injury was similar to that of mild hypothermia on rats with acute brain injury.
