BackgroundCervical cancer is the second most common gynecological cancer worldwide[1].86%of cervical cancer occurs in low-and middle-income countries,where cervical cancer mortality accounts for 88%of global cervical cancer mortality[2].Integration and persistent infection of human papillomavirus?HPV?at the junction of squamous epithelium and columnar epithelium?transformation region?of cervix is an important factor leading to cervical cancer.The transformed region is likely to become a reproducible malignant cancer stem cell?CSCs?.Recognition and targeting of cancer stem cells by tumor markers can early detect and cure cervical cancer[3].Cancer stem cells?CSCs?refer to a group of cancer cells that can proliferate indefinitely,self-renew and differentiate in many directions,and produce different phenotypes.They are composed of heterogeneous cell populations.Some cells in cancer tissues have stem cell characteristics.Tumor stem cells play an important role in tumor metastasis,recurrence and drug resistance.At present,CSCs markers and functional analysis are used to purify,identify and treat CSCs in vitro and in vivo.CD44 is a multifunctional transmembrane glycoprotein.It is an important member of the cell adhesion molecule family?CAM?.It was originally considered to be the receptor of hyaluronic acid?HA?.It is mainly responsible for cell-to-cell,cell-to-matrix adhesion.It is also an important marker of cancer stem cells[4].However,there are many variants of CD44,but the main subtypes of CD44 expression in cervical cancer and whether CD44 is a stem cell marker of cervical cancer are still controversial.In this paper,the expression of CD44 variants in cervical cancer cell lines and clinical specimens was detected and analyzed.ObjectiveTo study and analyze the variants of stem cell marker CD44 in cervical cancerMethods?1?qRT-PCR was used to detect the transcriptional level of various variants of CD44 in SiHa,HeLa and ME-180 cell lines.?2?The expression of CD44 in cervical cancer cell lines was further confirmed by gel electrophoresis.Western blot explored the protein level of CD44v6 in cervical cancer cell lines.?3?The expression levels and location of CD44v6 protein in 30 cases of cervical cancer tissues were detected by immunohistochemistry and analyze the correlation with clinical features and prognosis.?4?Using kaplan meier-plotter curve RNAseq database to analyse the prognosis of other common cancer stem cells markers ABCG2,SOX2 and KLF4 in cervical squamous cell carcinoma.Results?1?The SiHa,HeLa mainly expressed CD44s and CD44v6 variants,the ME-180 cell line mainly expressed CD44v variants,especially CD44v6.?2?The expression of CD44v6 in cervical squamous cancer tissues were positive,mainly in the cell membrane and cytoplasm.?3?The strong expression of CD44v6??9 points?was related to the stage of cervical cancer,lymph node metastasis and prognosis?P<0.05?,but it was not related to the degree of differentiation?P>0.05?.?4?ABCG2,SOX2 and KLF4 in cervical squamous cell carcinoma were negatively correlated with prognosis by using kaplan meier-plotter curve,while Lrig,BMI1,NANOG and UTF1 were not correlated with prognosis.Conclusion?1?Cervical cancer cell lines and tissues mainly express CD44v6 variant,SiHa and HeLa mainly express CD44s,ME-180 cell line mainly express CD44v;?2?High expression of CD44v6??9 points?in the invasion and metastasis of cervical cancer is an important biological marker,judging the prognosis of cervical cancer;?3?Survival analysis of ABCG2,SOX2 and KLF4 in cervical cancer stem cells may be prognostic markers by using kaplan meier-plotter curve.
