Chapter 1 The effect of non-surgical periodontal therapy on renal function in obese rats with experimental periodontitisObjectiveTo investigate the effect of non-surgical periodontal therapy on renal function inobese rats with experimental periodontitis.MethodsThirty-two SD rats were randomly divided into healthy group,obesity group,obesity+periodontitis group and periodontal therapy group.The serum creatinine,urea nitrogen and the early renal damage biomarker KIM-1 were detected.Results(1)The serum creatinine levels in obesity group and obesity+periodontitis group were significantly higher than that in healthy group(P=0.002,0.000).In periodontal therapy group,serum creatinine was lower than that in obesity group(P=0.073)and was significantly lower than that in obesity+periodontitis group(P=0.001).(2)The serum KIM-1 level in obesity+periodontitis group was the highest among four groups(P<0.05),in periodontal therapy group,it was lower than that in obesity group(P=0.346)and was significantly lower than that in obesity+periodontitis group(P=0.002).ConclusionNon-surgical periodontal therapy can effectively improve renal dysfunction caused by the compound factors of obesity and experimental periodontitis in rats.Chapter 2 The effect of non-surgical periodontal therapy on renal histopathology in obese rats with experimental periodontitisObjectliveTo investigate the effect of non-surgical periodontal therapy on renal histopathology in obese rats with experimental periodontitis.MethodsParaffin sections were made.HE,PAS,Masson trichrome staining were carried out and the electron microscopy specimens were performed.The renal histopathology was observed by optical and electron microscopy.Results(1)Inoptical microscopy,it showed vacuolar degeneration in obesity group.In obesity+periodontitis group,vacuolar degeneration aggravated and the loss of brush border and the movement of several nuclei of renal tubules were observed,which were much milder than that in periodontal therapy group.(2)Inelectron microscopy,vacuolar degeneration and the loss of microvilli of renal tubules were showed in obesity+periodontitis group.There was no difference in the basement membrane,renal interstitium and mitochondrion among four groups.ConclusionNon-surgical periodontal therapy can improve early destruction of renal structures in rats resulted from the compound factors of obesity and experimental periodontitis.Chapter 3 The effect of non-surgical periodontal therapy on serum inflammation,renal inflammation and oxidative stress in obese rats with experimental periodontitisObjectiveTo investigate the effect of non-surgical periodontal therapy on serum inflammation,renal inflammation and oxidative stress in obese rats with periodontitis.MethodsSerum LPS and IL-1? of rats were tested.TLR4 and NF-?B immunohistochemical staining were performed,qRT-PCR and western blot were used to detect the mRNA and protein levels of TLR4 and NF-?B in renal tissues.The oxidative stress indicators including MDA,8-OHdG,GSH and SOD were detected by ELISA and chemiluminescence.Results(1)There were no significant differences of serum LPS and IL-1? among four groups(P>0.05).(2)TLR4 and NF-?B were mainly expressed in renal tubular epithelial cells in immunohistochemistry,especially in obesity+periodontitis group.(3)The mRNA expression of TLR4 in obesity+periodontitis group was the highest among four groups.Theprotein expression of TLR4in obesity+periodontitis wassignificantly higher than that in healthy group and obesity group(P<0.001),and in periodontal therapy group,it was decresed than that in obesity group(P=0.285)and was significantly lower than that in obesity+periodontitis group(P<0.001).The mRNA and protein levels of NF-?B in obesity+periodontitis group were the highest among four groups,especially than that in obesity group and periodontal therapy group(P<0.05).(4)MDA and 8-OHdG in obesity+periodontitis group were the highest,and the SOD and GSH were the lowest among four groups(P<0.05).The MDA and 8-OHdG were decreased and the GSH was increased significantly inperiodontal therapy group than that in obesity+periodontitis group(P<0.05).ConclusionNon-surgical periodontal therapy can reduce the levels of renal inflammation and oxidative stress caused by obesity and experimental periodontitis in rats.
