| ObjectiveTo investigate the relationship among myosin phosphatase-targeting subunit I,(MYPT1)、regulatory light chain(RLC)and nonmuscle myosin heavy chain IIA(NMHCIIA)and their clinicopathological significance in breast invasive ductal carcinoma(IDC),the expressions of the three molecules in hyperplasia,fibroadenoma and IDC of breast were detected.It may provide new ideas and targets for clinical diagnosis and treatment of IDC.MethodsImmunohistochemistry(IHC)staining was used to detect the expression of MYPT1,RLC and NMHCIIA in hyperplasia,fibroadenoma and IDC of breast.SPSS v23.0 was used to analysis the correlation between the three molecules and several clinicopathological factors separately.ResultsIHC staining results showed that in hyperplasia and fibroadenoma of breast,MYPT1 is highly expressed in the glandular duct,but lowly in IDC.On the other hand,RLC was detected in glandular duct in both hyperplasia and fibroadenoma of breast.Moreover,the expression level of RLC is significantly increased in IDC compared to the other two lesions.NMHCIIA is expressed in all of the three types of lesions,among NMHCIIA has the highest expression in IDC.Statistic results showed that the expression levels of RLC and NMHCIIA were closely correlated in IDC.MYPT1 was negatively correlated with RLC or NMHCIIA expression levels.The expressions of MYPT1,RLC and NMHCIIA were correlated with age,TNM staging and 5-year survival rate of IDC patients.ConclusionsThe expression levels of MYPT1,RLC and NMHCIIA were closely related to IDC.MYPT1-RLC-NMHCIIA regulating pathway may plays an important role in occurrence and development of IDC,it may serve as a novel target of IDC therapy in clinic.In IDC,the expressions of MYPT1,RLC and NMHCIIA were clinically correlated with multiple clinicopathological factors and prognosis.To MYPT1,RLC and NMHCIIA,double-combined test or single test may be a critical reference for TNM staging and prognosis evaluation of IDC. |
