Background:Stroke is a multifactorial disease,the interaction of environmental factors and genetic factors plays an important role in the pathogenesis of stroke.There are about 15 million stroke patients in the world every year,of which 20% are fatal and more than 50% are cripple.Ischemic stroke accounts for more than 85% of stroke,and its proportion in China is increasing year by year,and there is a trend of younger.Single nucleotide polymorphism is the most common type of genetic variation.Recent advances in molecular genetics have made people realize that single nucleotide polymorphism is closely related to stroke.Superoxide dismutase(SOD)is the only known enzyme that scavenges free radicals directly and is essential for cell protection.A large number of animal experiments have shown that SOD is closely related to stroke.Histone Deacetylase 9(HDAC9)gene regulates gene expression through chromatin structural modification,but does not change DNA sequence.HDAC9 has been identified as the strongest risk gene for macrovascular stroke so far.However,the correlation between the polymorphisms of SOD2(rs2842960),SOD3(rs7655372),HDAC9(rs723296)and stroke remains unclear.The purpose of this study is to understand the genetic risk factors of stroke in DaLi and to provide scientific basis for deciphering its basic mechanism,promoting the identification of new therapeutic targets and optimizing preventive strategies.Objective: To study the correlation between polymorphisms of SOD2(rs2842960),SOD3(rs7655372),HDAC9(rs723296)and stroke patients in DaLi.Methods: 155 stroke patients and 128 healthy persons of Han were randomly selected by case-control method.Real-time PCR was used to amplify DNA and genotyping of each SNP by modified TaqMan Fluorescence Quantitative Technology.The traditional risk factors of stroke were analyzed by T-test,Chi square test and logistic regression statistics.The genetic balance was tested by Hardy-Weinberg equilibrium law.Logistic regressionanalysis corrected the traditional risk factors and analyzed the influence of SNP polymorphism on susceptibility to stroke.The correlation between rs723296 polymorphism and lipid levels was analyzed by linear regression analysis.Multivariate dimensionality reduction was used to analyze the effect of gene-gene interaction on stroke..Results:(1)Traditional risk factors: Blood glucose(p=0.00 OR=2.885,95%CI 1.86-4.41).Cholesterosterol(p=0.00 OR=2.378,95%CI 1.55-3.66).Triglycerides(p=0.013 OR=1.657,95%CI 1.11-2.47).LDL(p=0.013 OR=1.641,95%CI 1.11-2.42).Leukocyte(p=0.013 OR=1.641,95%CI 1.11-2.42).Systolic pressure(p=0.03 OR=1.017,95%CI 1.00-1.03).Erythrocyte(p = 0.007 OR = 0.477,95% CI 0.28-0.82).Their p values are less than 0.05.(2)The dominant model of SOD2(rs2842960),CC/(CT+TT): p=0.383,OR=0.767,95%CI:0.423-1.392.(3)The additive model of SOD3(rs7655372),GG/GA: p=0.028,OR=3.188,95%CI: 1.134-8.962.(4)The additive model of HDAC9(rs723296),TC/TT: p=0.021,OR=2.669,95%CI: 1.046-4.923.(5)The polymorphisms of HDAC9(rs723296)and lipid levels in the experimental group shows: LDL(β=0.171,p=0.456),cholesterol(β=-0.141,p=0.502),triglyceride(β=0.035,p=0.865);in the control group: LDL(β=0.364,p=0.301),cholesterol(β=-0.573,p=0.083),triglyceride(β=0.340,p=0.337).Their p values are greater than 0.05,without statistical significance.(6)There was strong interaction among SOD2(rs2842960),SOD3(rs7655372)and HDAC9(rs723296),but there was no significant effect on the onset of stroke(p > 0.05).Conclusion: Polymorphisms of SOD3(rs7655372)and HDAC9(rs723296)increase the risk of stroke in DaLi.Polymorphisms of SOD2(rs2842960)are not associated with susceptibility to stroke.The polymorphism of the HDAC9(rs723296)was not associated with lipid level.The gene-gene interaction of SOD2(rs2842960),SOD3(rs7655372)and HDAC9(rs723296)had no significant effect on the onset of stroke. |
