Objective:The study is aimed at examining the expression of Rel B/My D88 in PBMC and IL-39 in the serum of patients with neuromyelitis optica spectrum disorders.Then we explore the correlation between the expression levels of Rel B,My D88 and IL-39 with NMOSD disease severity.Subjects and Methods:In the first part,we recruited 21 patients with NMOSD patients and 16 healthy controls to examine the expression of Rel B and My D88 protein in PBMC.These NMOSD patients were admitted to the Department of Neurology and the Neuroscience Center at The First Hospital of Jilin University from April 2018 to February 2019.In the second part,we recruited 50 patients with NMOSD patients,30 patients with non-inflammatory neurological disorders(NND)and 78 healthy controls to examine the expression of IL-39 in the serum.These NMOSD patients were admitted to the Department of Neurology and the Neuroscience Center at The First Hospital of Jilin University from November 2016 to February 2019.All NMOSD patients were diagnosed by two neuroimmunological physicians and met the 2015 new diagnostic criteria.Also,we assessed the severity of these NMOSD patients by using the Expanded Disability Status Scale(EDSS)and collected the clinical characteristics of these patients.All the subjects provided written informed consent.1.The expression levels of Rel B and My D88 in PBMC of NMOSD patients and healthy controls were tested by using the Western Blot.2.The expression levels of serum IL-39 in NMOSD patients,NND patients and healthy controls were tested by using the enzyme-linked immunosorbent assay(ELISA).3.To analyze the correlation between Rel B,My D88 and IL-39 levels and the EDSS score as well as ARR.Results:1.The expression of Rel B in PBMC of NMOSD patients was significantly lower than that of healthy controls(P=0.0019).2.The expression of My D88 in PBMC of NMOSD patients was significantly higher than that of healthy controls(P=0.0471).3.The expression of Rel B and My D88 in PBMC of NMOSD patients had no significant correlation with EDSS(P=0.3532,P=0.1782)and ARR(P=0.3077,P=0.7141).4.The serum IL-39 levels in patients with NMOSD were significantly higher than that in NND patients and healthy controls(P=0.0352,P<0.0001).5.The serum IL-39 levels in NMOSD patients were positively correlated with EDSS(P=0.0337),and had no significant correlation with ARR(P=0.9150).Conclusion:1.The Rel B expression was significantly lower in PBMC of NMOSD patients;2.The My D88 expression was significantly higher in PBMC of NMOSD patients;3.The expression of Rel B and My D88 in PBMC of NMOSD patients had no significant correlation with EDSS and ARR;4.The serum IL-39 levels were significantly higher in NMOSD patients.5.The serum IL-39 levels in NMOSD patients were positively correlated with EDSS and had no significant correlation with ARR.
