| Objective:To study the therapeutic effect and possible mechanism of penehyclidine hydrochloride combined with lipoic acid in the treatment o f lung injury induced by paraquat poisoning in rats.Methods: Two hundreds male SD rats were randomly divided into 5 gro ups: blank control group(group A)and experimental group(group B,C,D,E).Except the blank control group,the rats in the other groups were in jected intraperitoneally with 20% paraquat solution 20mg/kg(diluted wit h normal saline to 1ml injection)to establish the paraquat poisoning mod el of SD rats.The blank control group was injected intraperitoneally with the same amount of normal saline.30 minutes after being injected Paraqu at,the rats in the experimental groups were intraperitoneally injected with different substances: penehyclidine hydrochloride group(0.1mg/ kg(B group),lipoic acid 100mg/kg(group C),lipoic acid 100mg/kg combined with penehyclidine hydrochloride 0.1mg/ kg(D group)and equal volume saline(group E).Until the end of this experiment,the rats in experimental groups were repeatedly injected every 24 hours.Rats in every group were randomly divided into four subgroups on the 1st,7th,14 th and 28 th day,with 10 rats in every group.On the 1st,7th,14 th and 28 th day of the experiment,the animals were killed and different indicators were examined,including the level of IL-6,TNF-α in BALF,the pathomorphological changes of lung tissue,the expression of transforming growth factor TGF-β 1 in lung tissue and the level of TGF-β 1 in blood.Results: The level of IL-6 and TNF-α in the lung tissues and TGF-β1 in the blood of rats poisoned with PQ(B,C,D,E)were significantly higher than those in the control group(A)at all time points(P<0.05).On the 7th,14 th and 28 th day,the level of each indicator in groups B,C and D was significantly higher than that in group E(P<0.05).On the 14 th and 28 th day,the level of each indicator in the D group was lower than that in the B and C groups,which was statistically significant(P<0.05).Conclusion: Both penehyclidine hydrochloride and lipoic acid can alleviate acute lung injury and inhibit interstitial fibrosis in paraquat intoxicated rats. |
