Analysis Of Protein And Antibody Of MAGE-D4 In Glioma Patients’ Sera And Its Clinical Significance | | Posted on:2019-12-18 | Degree:Master | Type:Thesis | | Country:China | Candidate:H Tan | Full Text:PDF | | GTID:2404330575462841 | Subject:Neurosurgery | | Abstract/Summary: | PDF Full Text Request | | Objective:To detect sera glioma-associated antigen MAGE-D4 protein and antibody of glioma patients.Than the clinical significance was analyzed,and the feasibility that MAGE-D4 is use for diagnosis and prognosis of glioma was Evaluated preliminarily,which make a foundation for the clinical application of MAGE-D4.Methods:(1)Clinical specimens: 124 cases of Intracranial tumors patients in hospitalized from 2013 to 2015 which was treated surgically were collected,there is 92 cases of gliomas(including 46 cases of astrocytoma,9 cases of oligodendroglioma,25 cases of glioblastoma,10 cases of medulloblastoma,2cases of ependymoma)and 32 cases of other intracranial tumors(including 13 cases of meningiomas,2 cases of Intracranial metastases,11 cases of pituitary adenomas,6 cases of schwannoma);(2)patient’s sera MAGE-D4 protein was detected using Commercial sandwich ELISA kit,and the correlation between sera MAGE-D4 protein and clinical parameters was analyzed.(3)In our research group,the MAGE-D4 recombinant truncated protein that had been prepared wascoated,which Create a way to detect sera MAGE-D4 antibody OD value of Healthy volunteers and gliomas using Indirect ELISA assay.Than the correlation between sera MAGE-D4 antibody OD value and clinical parameters was analyzed.(4)The detection of sera MAGE-D4 protein and antibody was combined,and according to the result,ROC curve and the OS curve of glioma could be described.Results:(1)Serum MAGE-D4 protein concentrations of glioma patients were significantly higher than healthy volunteers and other intracranial tumor patients(P < 0.001);There was not different serum MAGE-D4 protein concentrations from high-grade glioma and low-grade glioma patients(P> 0.05);There was also not differentserum MAGE-D4 protein concentrations in various histological types of glioma patient(P> 0.05);Serum MAGE-D4 protein of Intracranial tumors was assessed:The total positive rate of serum MAGE-D4 protein was 22.58%(28/124)in various intracranial tumors,and the positive rate of MAGE-D4 protein in glioma was 27.71%(25/92),the positive rate of Other intracranial tumors were 9.38%(3/32);(2)Serum MAGE-D4 antibody OD value of glioma patients were significantly higher than healthy volunteer and other intracranial tumor patients(P < 0.001);There was not different Serum MAGE-D4 antibody OD value from high-grade glioma and low-grade glioma patients(P> 0.05);There was also not different serum MAGE-D4 antibody OD value in various histological types of glioma patient(P> 0.05);Serum MAGE-D4 antibody OD value of Intracranial tumors was assessed:The total positive rate of Serum MAGE-D4 antibody OD value was 34.7%(43/124)in various intracranial tumors,and the positive rate of Serum MAGE-D4 antibody OD value of glioma was 42.4%(39/92),the positive rate of Serum MAGE-D4 antibody OD value of Other intracranial tumors were 12.5% ??(4/32);(3)Comprehensive analysis of MAGE-D4 protein and antibody:The positive rate of serum MAGE-D4 protein or antibody of glioma patients was 58.7%.MAGE-D4 is apotential glioma serum biomarker.(4)Prognosis analysis: Neither proteins nor antibodies,The over survival of positive and negative MAGE-D4 glioma patients showed no significant difference(P>0.05).Conclusion:Both the serum MAGE-D4 protein concentration and antibody OD value of glioma patients are higher than healthy volunteers and other intracranial tumor patients;MAGE-D4 protein and antibody have no obvious correlation with the prognosis ofglioma patients;serum MAGE-D4 protein and antibody are potential glioma serum biomarker,which`s Clinical significance worth to be further explored. | | Keywords/Search Tags: | MAGE-D4, glioma, Serum biomarkers, ELISA | PDF Full Text Request | Related items |
