Autophagy In Endometrial Hyperplasia After Autogenous Arteriovenous Fistula Surgery In Rabbits

Objective:To investigate the relationship between intimal hyperplasia and autophagy after autogenous arteriovenous fistula(AVF).Methods: Thirty male New Zealand rabbits were randomly divided into 3groups,10 in each group(experiment I,II,III).In the experimental group(n=30),the left external jugular vein and the common carotid artery were dissected and the end-to-side anastomosis was performed.The right blood vessels of the experimental group were used as the control group(control group I,group II,group III),and the right external jugular vein and common carotid artery were separated but not matched.At the 1st,2nd,and 4th week after the operation,a section of the external jugular vein of the arteriovenous anastomosis vein in the experimental group I,II,and III was taken,and the control group took the corresponding position of the vein.Measurement of the thickness of the external jugular vein intima and the expression of detection proteins(Beclin-1,LC3-II,P62).Results:1.Intima thickness: There was no difference in intimal thickness of the control group(P>0.05);the intima of the experimental group were thickened(P<0.05),and the intima thickness increased gradually from the experimental group I to the third group(P<0.05).2.Protein expression: There was no difference in the expression of Beclin-1,LC3-II and P62 in the control group(P>0.05).The expressions of Beclin-1 and LC3-II in the experimental group were up-regulated(P<0.05),and from experiment I.The expression of group I to group III was gradually increased(P<0.05),and the expression of P62 was down-regulated(P<0.05),and its expression showed a downward trend(P<0.05).3.Correlation analysis: There was a positive correlation between Intima thickness and Beclin-1 and LC3-II expression(r=0.887,P=0.000;r=0.919,P=0.000),and negative correlation with P62(r=-0.788,P =0.000).Conclusion:1.Autophagy is activated up-regulation and intimal hyperplasia occurs after AVF,autophagy participates in the process of intimal hyperplasia after AVF;2.With the prolongation of AVF exposure time,autophagy expression was positively correlated with intimal hyperplasia,suggesting a causal relationship between the two.
