ObjectiveTo explore whether SOX9 can be used as a predictor of progesterone response and recurrence in endometrial hyperplasia and its clinical significance.Methods72 cases of surgical patients from the Fourth Affiliated Hospital of Guangxi Medical University from January 2011 to January 2016 were collected,including 31 cases of endometrial hyperplasia without atypia(EH),21 cases of typical hyperplasia(AH),and normal endometrial tissue containing 10 cases of secretory endometrium(SE),10 cases of proliferative endometrium(PE).Among them,31 cases of EH and 11 cases of AH cases were treated with progesterone.Immunohistochemistry was used to detect the expression of SOX9in SE,PE,EH and AH,and the difference of SOX9 expression before and after progesterone treatment.The clinical and pathological characteristics such as age,number of pregnancies and deliveries,menopause or not,body mass index(BMI)and recurrence were analyzed.(1)The expression of SOX9 in SE,PE,EH and AH showed an increasing relationship(P<0.05).(2)SOX9 was decreased in each group after progesterone treatment(P<0.05),and The expression of SOX9 in the effective group of progesterone treatment was significantly decreased,and the difference was statistically significant(P<0.05);the expression of SOX9 in the ineffective group was not significantly decreased,and the difference was not statistically significant(P>0.05).(3)The expression of SOX9 was not significantly correlated with the recurrence time of patients with endometrial hyperplasia.Cox analysis showed that BMI>25 and Norethisterone(NET)were independent factors affecting the recurrence of patients with endometrial hyperplasia.Conclusions(1)The expression of SOX9 in SE,PE,EH and AH was increased gradually,which could be related to the occurrence and development of endometrial hyperplasia.(2)The down-regulation of SOX9 can be used as a reference index to predict the efficacy of progesterone therapy.(3)BMI>25kg/m~2 and NET were risk factors for recurrence in patients with endometrial hyperplasia.
