Background Lung cancer is a common respiratory disease with high morbidity and mortality.It consists of small cell lung cancer(SCLC)and non-small cell lung cancer(NSCLC),of which NSCLC is the most common type,accounting for 80%~ 85% of the total lung cancer.The importance of lung cancer is gradually increasing at home and abroad,but the rates of misdiagnosis and missed diagnosis are still high,and one of the important reasons is the lack of effective diagnostic methods.Imaging examination and bronchoscopy are the main two methods to diagnose lung cancer.Imaging test limits its use because of the high cost,potential radiation and allergic reactions.The use of bronchoscopy is also limited,because it often leads to complications,such as nosebleed,laryngeal edema,and spontaneous pneumothorax.Therefore,finding an effective,affordable and non-invasive detection method is the most important diagnostic and treatment strategy for lung cancer.In recent years,more and more studies have shown that non-coding RNA(nc RNA)plays an important regulatory role in the occurrence and development of tumors.It includes long non-coding RNAs(lnc RNAs),micro RNAs(mi RNAs),transcribed pseudogenes and circular RNAs(circ RNAs),which are closely related to biological behaviors of tumor cells,such as proliferation,metastasis,invasion,and apoptosis.Recently,more and more studies have shown that lnc RNA plays an important regulatory role in the development of tumors.However,there are few studies on lnc RNA NR2F2-AS1,especially in non-small cell lung cancer.In this study,we detected the expression level of lnc RNA NR2F2-AS1 in non-small cell lung cancer tissues and cells,and further explored the specific mechanism of lnc RNA NR2F2-AS1 in NSCLC cells.This study consists of two parts.The first part is the expression of lnc RNA NR2F2-AS1 in lung cancer tissues and lung cancer cells.The second part is the effect of disregulated lnc RNA NR2F2-AS1 on the proliferation,invasion and apoptosis of lung cancer cells.Part? The expression of lnc RNA NR2F2-AS1 in non-small cell lung cancer tissues and cellsObjective The expression of lnc RNA NR2F2-AS1 in non-small cell lung cancer tissues and cell lines was detected.The correlation between lnc RNA NR2F2-AS1 and the clinicopathological characteristics of non-samll cell lung cancer patients was further evaluated.Method1 Specimens were collected,including 39 cases of lung cancer and corresponding normal tissues.2 The expression of lnc RNA NR2F2-AS1 in lung cancer tissues and cell lines was detected by q RT-PCR.3 According to the expression level of lnc RNA NR2F2-AS1,we carried out the statistical analysis in the field of age,gender,tumor TNM stage and lymph node metastasis.Results1 Compared with adjacent tissues,the expression of lnc RNA NR2F2-AS1 was significantly higher in lung cancer tissues(P<0.05).2 Compared with human normal bronchial epithelial cells(NHBE),lnc RNA NR2F2-AS1 was highly expressed in lung cancer cells(P<0.05).3 The expression of lnc RNA NR2F2-AS1 was remarkably associated with the TNM stage(P=0.029)and the status of lymphatic metastasis of patients(P=0.015).And there was no relationship between the expression of lnc RNA NR2F2-AS1 with age(P=0.177)and gender(P=0.752).Part? Downregulated lnc RNA NR2F2-AS1 on the Proliferation,Invasion and Apoptosis of lung cancer cellsObjectiveThe effects of downregulated lnc RNA NR2F2-AS1 on the proliferation,invasion and apoptosis of A549 and SPC-A-1 cells was analyzed.And we also preliminarily evaluated the possible molecular mechanism of lnc RNA NR2F2-AS1 in lung cancer.Methods 1 Si-lnc RNA NR2F2-AS1 was designed by using si RNA interference technology,and q RT-PCR was used to detect the expression of lnc RNA NR2F2-AS1 in A549 and SPC-A-1 cells.2 CCK-8 assay was applied to explore the proliferation of A549 and SPC-A-1 cells.3 Transwell assay was performed to detect the invasion of A549 and SPC-A-1 cells.4 Flow cytometry and Caspase 3/7 activity detection was used to analyze the apoptosis of A549 and SPC-A-1 cells.Results1 Si-lnc RNA NR2F2-AS1 was respectively transfected into A549 and SPC-A-1 cells,the results of q RT-PCR showed that the expression of lnc RNA NR2F2-AS1 was successfully inhibited through the transfection(P<0.05).2 The result of CCK-8 assay indicated that the proliferative ability of A549 and SPC-A-1 cells in the si-lnc RNA NR2F2-AS1 group was significantly decreased(P<0.05)compared with the si-NC group.3 The result of Transwell assay indicated that the invasive ability of A549 and SPC-A-1 cells in the si-lnc RNA NR2F2-AS1 group was significantly decreased(P<0.05)compared with the si-NC group.4 The result of Flow cytometry and Caspase 3/7 activity detection assay indicated that the apoptostic ability of A549 and SPC-A-1 cells in the si-lnc RNA NR2F2-AS1 group was statistically increased(P<0.05)compared with the si-NC group.Conclusion1 The expression of lnc RNA NR2F2-AS1 was significantly higher in NSCLC tissues and lung cancer cell lines and it was related to TNM stage and lymphatic metastasis of NSCLC patients.2 Downregulating the expression level of lnc RNA NR2F2-AS1 contributed to the inhibition of proliferation and invasion and promotion of apoptosis in NSCLC cells.
