Head and neck cancer is one of the types of tumors that threaten human life.Throat squamous cell carcinoma is one of the common pathological types.In recent years,the incidence of laryngeal squamous cell carcinoma has increased year by year,and the cause of the disease is the result of multi-factor synergy.Recently,a variety of methods have been used in combination in its clinical treatment,including surgery and radiotherapy,but the treatment effect has not been significantly improved,and the patient's quality of life is seriously affected after surgery.In recent years,many scholars have studied the pathogenesis of laryngeal cancer at the molecular level in order to find more effective treatment methods.MiRNAs are small,non-coding,double-stranded RNA molecules that mediate the expression of a target gene with complementary sequences.Different scholars have discovered the relationship between many miRNAs and the occurrence and development of diseases,and there are also many abnormal expressions of miRNAs in laryngeal cancer tissues.However,little is known about the molecular mechanism of miR-384 in laryngeal cancer.WISP1,a Wnt-induced secreted protein,is part of the CCN family and plays an important role in various tumor tissues.In this study,we set out to analyze the expression levels of miRNA-384 and WISP1 proteins in tissue cells and correlate the results with the clinical subtypes of patients.And statistical analysis of the relationship between the two,provide a theoretical basis for the subsequent exploration of new therapeutic targets for laryngeal cancer.Objective1 The expression levels of miRNA-384 and WISP1 proteins in laryngeal squamous cell carcinoma and adjacent normal tissues were detected respectively.2 To study the relationship between the expression levels of miRNA-384 and WISP1 proteins in laryngeal squamous cell carcinoma and clinical subtypes.3 The correlation between MiRNA-384 and WISP1 protein expression was analyzed.MethodsReal-time quantitative PCR?Q-PCR?was used to detect the expression of miRNA-384 in laryngeal squamous cell carcinoma and adjacent normal tissues.U6was used as the internal reference gene,and the target gene was semi-quantified by2-??Ct method.Immunohistochemical?IHC?was used to detect the expression of WISP1 protein in laryngeal squamous cell carcinoma and adjacent normal tissues.SPSS 25.0 statistical software was used to analyze the relationship between the expression of miRNA-384 and WISP1 protein in laryngeal squamous cell carcinoma and the clinical subtypes,and the correlation between miRNA-384 and WISP1protein was analyzed statistically.Results1 The relative expression of miRNA-384 in laryngeal squamous cell carcinoma was lower than that in adjacent normal tissues?P<0.05?,and the difference was statistically significant.The expression of WISP1 protein in laryngeal squamous cell carcinoma was higher than that in adjacent normal tissues?P<0.05?,and the difference was statistically significant.2 The expressions of miRNA-384 and WISP1 in laryngeal squamous cell carcinoma were not related to age,sex,differentiation and clinical stage?P>0.05?.The expression of miRNA-384 in patients with positive lymph node metastasis was lower than that in negative patients?P<0.05?,but the expression of WISP1 protein was higher than that in negative patients?P<0.05?.The difference was statistically significant.3 The correlation between miRNA-384 and WISP1 protein expression in laryngeal squamous cell carcinoma was negatively correlated by contingency table correlation analysis.?r=-0.899,P<0.05?.Conclusions1 The results showed that miRNA-384 was lowly expressed in laryngeal squamous cell carcinoma,and WISP1 protein was highly expressed in laryngeal squamous cell carcinoma.2 The results suggest that the low expression of miRNA-384 and the high expression of WISP1 protein are related to the lymphatic metastasis of laryngeal squamous cell carcinoma,which is helpful to assess the severity of the disease.3 MiRNA-384 and WISP1 proteins have a negative correlation in the occurrence and evolution of laryngeal squamous cell carcinoma,which provides a theoretical basis for the specific pathogenesis of laryngeal squamous cell carcinoma,and can also provide a gene-targeted therapy for laryngeal squamous cell carcinoma,which is a prospective treatment idea.
