Objective: The aim of this study is to investigate the effect of NOX1 protein on epithelial-mesenchymal transition(EMT)of ovarian cancer cell,A2780 cell and SK-OV-3 cell and better understand the mechanism of tumorgenesis.Methods: 1 Transient transfection of NOX1-expressing plasmids was used to overexpress NOX1 protein in A2780 cell and SK-OV-3 cell;2 Lentivirus-mediated RNA interference(RNAi)was employed to establish NOX1 stable knockdown cells;3 Clone forming assay was carried out to assess the ability to clone formation both of control cells and NOX1 knockdown cells;4 MTT analysis was employed to analyzed the effect of NOX1 stable knockdown on A2870 cell and SK-OV-3 cell proliferation;5 Scratch test were employed to study the effect of NOX1 knockdown on migration;5 Transwell chamber assay were employed to assessed the migration and invasion of A2780 cell and SK-OV-3 cell;7 Western blot was used to detect the expression of EMT markers in NOX1 overexpression cells and NOX1 stable knockdown cells.Res?lt: Stable knockdown of NOX1 significantly suppressed the ability to clone transformation,proliferation,cell migration and invasion,both in A2780 cells and SK-OV-3 cells(p all<0.05);western blot analysis revealed an elevated level of N-cadherin and vimentin and a reduced level of E-cadherin in NOX1-overexpressing cells,but a reduced level ofN-cadherin and vimentin and an elevated level of E-cadherin in NOX1 stable knockdown cells(p all<0.05).Conclusion: NOX1 significantly promotes the EMT of ovarian cancer A2780 cells and SK-OV-3 cells.
