Objective: To explore the effects of procyanidins on the improvement of cognitive function in APP/PS1 double-transgenic AD model mice and discuss its possible mechanism of mitochondrial respiratory function and oxidative stress state.Methods: The animals were divided into three groups: Control(20 C57BL/6 mice),AD model(20 APP/PS1 mice),AD+PC intervention(20 APP/PS1 mice).The intervention began at 2 months of age and lasted for 5 months.At the age of 7 months,Morris water maze was used to evaluate the changes in cognitive ability,and oxygraph-2k mitochondrial respiration instrument was used to evaluate the mitochondrial function of the hippocampus.The levels of SOD,CAT,GSH-PX,GSH and MDA in brain tissues were determined by the kit method.Results: The procyanidins can improve the ability of spatial learning and memory function in APP/PS1 mice.In place navigation test,the escape latency of the AD+PC group was significantly shortened compared with the AD model group(p<0.05),and there was no significant difference in the swimming speed and distance between the groups.In spatial probe test,the target quadrant travel distance of AD+PC group was significantly increased compared with AD group(p<0.05),but there was no significant improvement in the first time of crossing the platform,target quadrant stay time,target quadrant occurrence times,crossing the platform times and target quadrant travel distance.Procyanidins intervention improved the hippocampal mitochondrial CIIETS respiratory function in the APP/PS1 dual-transgenic model mice(p<0.05),but did not significantly improve the respiratory inhibition rate and mitochondrial oxidative phosphorylation respiratory function.The activity of CAT and GSH-PX in the brain tissue of the APP/PS1 dual-transgenic model mice was procyanidins by procyanidins(p<0.05),and assisted to improve the imbalance of the oxidation and antioxidant system in the mice.Conclusion: Procyanidins can improve the cognitive function of the APP/PS1double-transgenic AD model mice,improve the CIIETS respiration of mitochondria,and improve the activity of antioxidant enzymes in brain tissue to help improve theiroxidative stress status.
