Objective: To investigate the protective effects and its mechanism of procyanidins from the lotus seedpod on the learning and memory of APP/PS1 double transgenic dementia model mice.Methods: Forty two-month old APP/PS1 double transgenic dementia model mice were randomly divided into following groups: AD model group,LSPC group.Mice in LSPC group were administered with 100 mg/kg BW LSPC for 5 months.Meanwhile,20 wide C57/BL mice were considered as normal control group.The AD model group and normal control group were given equal volume of water.Morris water maze test was used to observe the difference of learning and memory function of mice;electrophysiological apparatus were used to measure the LTP amplitude;the expressions of ?-amyloid protein(A?),SIRT1,CREB/ pCREB,BDNF in hippocampus and cortex were quantified by western blot method.Results:(1)In Morris water maze task,LSPC can improve the function of spatial learning and memory in mice.Compared with AD model group,LSPC groups had significantly shorter escape latency during the training days,but the path length percent in platform quadrant and the crossing times across platform were significantly increased(P<0.05).(2)In electrophysiological recording experiment,the amplitude of LTP in AD model group was significantly decreased(P<0.05).But LSPC administration can significantly up-regulate the formation of LTP in hippocampus(P< 0.05).(3)In Western blot analysis,compared with control group,the content of A? was markedly increased,while the protein expression of,SIRT1,BDNF and the phosphorylation of CREB were decreased significantly in mice of APP/PS1 transgenic group(P<0.05).After LSPC intervention,the level of A? was significantly reduced compared with APP/PS1 transgenic group,the expression of SIRT1 and BDNF,the phosphorylation of CREB were significantly up-regulated(P<0.05).Conclusion: The potential neuroprotective role of LSPC in cognitive function is accomplished by enhancing LTP via mediating related protein expression and activity in APP/PS1 transgenic mice.
