Objectives:To study the pathogenic characteristics of hand-foot-mouth disease in Urumqi from 2013 to 2017,and to analyze the three distributions,pathogen composition,virus isolation and genetic evolution of EV71 local strains in hand-foot-mouth disease in Urumqi,providing data for prevention and control of hand,foot and mouth disease.Support and theoretical basis.Methods:The laboratory specimens of patients with hand,foot and mouth disease at various levels of medical institutions in Urumqi were collected from 2013 to 2017.The nucleic acid detection was carried out by real-time fluorescent PCR.The positive samples of nucleic acid detection were used for virus isolation and culture,and the specimens under different conditions were studied.The strain of hand,foot and mouth disease virus was obtained by the influence of the isolation rate of the strain.The RT-PCR was further detected and its type was identified.The full-length gene amplification and sequence detection of the capsid protein VP1 region of EV71 strain was performed.SPSS 20.0 was used.The software performs statistical analysis on laboratory test data.Results:1.Among the 3867 specimens,the positive rate of enterovirus was 76.44%(2956/3867),the male-female ratio was 1.49:1(2313/1554),the age was mainly concentrated under 6 years old(91.36%),and the popular time was 5-7.In the month(76.29%),the top three regions were the water area(752 cases),the new urban area(743 cases)and the sand area(734 cases).2.The best cell line for EV71 virus isolation and culture is RD cell line.The optimal culture temperature is 35?,the serum concentration is 2%,the inoculum size is 300ul,and the inoculation method is direct inoculation.The sample collection is best with fecal specimens.3.The EV71 local strain in Urumqi belongs to the C4a subtype.Conclusion:The positive rate of enterovirus in patients with hand,foot and mouth is still at a high level,and there are obvious differences in seasonal,population and geographical distribution characteristics.The dominant strains of hand,foot and mouth disease pathogens change alternately every year,causing different degrees of hand,foot and mouth disease epidemic.The EV71 local strain in Urumqi has high homology with other strains in China.
