Objective:Polygonati Rhizoma is a dried rhizome of the Liliaceae family Polygonatum,which is a traditional Chinese medicinal and edible plant.Polygonatum kingianum Coll.et Hemsl is one of the origins plants of Polygonati Rhizoma recorded in Chinese Pharmacopoeia,and it is named“Dahuang Jing”.As a Yunnan local medicinal herbs,Polygonatum kingianum is mainly produced in Yunnan,and it is famous for its excellent quality and large market share.Modern pharmacological studies have shown that polysaccharide from Polygonatum kingianum have the effect ofanti-fatigue and immune enhancement,and itit have no obvious side effects,so we chose the Polygonatum kingianum polysaccharide as raw material to preparation of buccal tablets,and preliminary studied on its anti-fatigue effect.Firstly,we compared the monosaccharide composition of polysaccharide from three origin sources of Polygonati rhizoma,and their effective of anti-fatigue was analyzed.After that,Polygonatum kingianum polysaccharide was decided as raw material.The extraction processing of polysaccharide from Polygonatum kingianum was optimized in this step.Secondly,Polygonatum kingianum polysaccharide buccal tablets'preparation process was determined,its anti-fatigue efficacy was studied too.The completion of the whole will further explore its efficacy in anti-fatigue and mechanism of the Polygonatum kingianum polysaccharide.This research would explain and demonstrate the medicinal value and health care effect of polysaccharide,which provide a preliminary basis for the development of an anti-fatigue health food.It also provided an important scientific basis for the further development and application of plant polysaccharide in anti-fatigue health field.Methods:Using pre-column derivatization of high-performance liquid chromatography analyzed the compositions of three origin sources of Polygonati rhizoma polysaccharide's monosaccharidecomponent,protein,total polysaccharide and uronic acid content.The weight-bearing swimming to exhaustive experiment compared anti-fatigue effects of three origin sources of Polygonati rhizoma polysaccharide;Four kinds of different processing methods were used to prepare four kinds of products,and the ideal starting sample was determined by the content of polysaccharides;L9(3~4)orthogonal test was used to optimize the extraction conditions.The dry tablet granules were used to develop the buccal tablets,according to the taste,appearance and tableting of the buccal tablet.The single factor study method was used to screen the formula of the buccal tablets and determine the best prescription;The buccal tablets were examined according to the requirements of the tablets in the people's republic of China pharmacopoeia;Studied the effect in anti-fatigue and anti-oxidation of the scutellaria baicalensis polysaccharide tablets by using the mouse weight-bearing swimming to exhaustive experiment.Results:The monosaccharide composition of three Polygonati rhizoma polysaccharide was:Polygonatum kingianum polysaccharide composed by 7 monosaccharidesmannose,ribose,rhamnose,galacturonic acid,glucose,galactose and arabinose,among them;Ribose,buckthorn Sugar and arabinose are first reported by this study.Both the Polygonatum sibiricum polysaccharide and Polygonatum cyrtonema were composed by 9 monosaccharides.Besides those mentioned above,another two monosaccharides are glucosamine hydrochloride and glucuronic acid.The proportion of polysaccharide was varies in three origin sources of Polygonati rhizoma.The percentages of polysaccharides in Polygonatum kingianum Coll.et Hemsl,Polygonatum sibiricum and Polygonatum cyrtonemawere 71.12%,71.8%and73.09%respectively.The protein contents of crude polysaccharides were 11.478%,10.63%and 13.688%respectively,and the uronic acid content was 15.01%.22.81%,18.11%.Results of anti-fatigue activity experiment revealed that the experimental groups of three origin sources of Polygonati rhizomapolysaccharides all have a certain tendency to prolong the swimming time of mice,which indicate that three polysaccharides have anti-fatigue effect.Based on prolonging the swimming time of mice,Polygonatum kingianum polysaccharide group was better than Polygonatum sibiricum polysaccharide group and Polygonatum cyrtonema polysaccharide group.Based on the measured data of blood lactate(LD),Polygonatum cyrtonema polysaccharide group was better than from Polygonatum kingianum polysaccharide group andPolygonatum sibiricum polysaccharide group.On the basis of liver glycogen(HG),Polygonatum sibiricum polysaccharide group was superior to Polygonatum kingianum polysaccharide group andPolygonatum cyrtonema polysaccharide group.According to experimental result,it was found that the extraction rate of fresh eucalyptus polysaccharide was the highest,so it was used as the extraction raw material,and extraction method is freshly squeezed.Finally,the optimal extraction process was:ration of liquid and material was 1:20,ultrasonic time was 50 min,the number of centrifugation was 3 times,and the alcohol content was 90%.The single-factor method is used to screen the formula of buccal tablets to determine the best prescription as 20%polysaccharides,50%lactose,15%mannose and 15%xylitol.Pharmacological experiments showed that the polysaccharides obtained could prolong the weight-bearing swimming time of mice.The high-dose group of Polygonatum kingianum polysaccharide can significantly reduce lactate dehydrogenase(LDH),muscle phosphokinase(CK),serum urea nitrogen(BUN),malondialdehyde(MDA),and polysaccharides.The low dose group of tablets could significantly reduce superoxide dismutase(SOD).Conclusion:The polysaccharide of Polygonati rhizoma has good anti-fatigue effective and abundant resources.The extraction method of Polygonatum kingianum polysaccharide is stable and feasible.According to the best prescription determined from the experiments,the buccal tablets of the Polygonatum kingianum polysaccharide have good mouth-feel and smooth surface.The tablet has anti-fatigue effect,and it would be a basis for development and application on functional health products of Polygonatum kingianum.
