Objective:To explore the experimental and theoretical studies of traditional Korean medicine Shuizhixiongdan powder(SZXDS)on therapeutic effect and its mechanism of cerebral ischemia reperfusion rats,and to provide theoretical basis for the clinical application and further experimental study of SZXDS.Method:The clean-grade SD rats were randomly divided into sham operation group,model group,low dose SZXDS group and high dose water SZXDS group.The suture method was used to conduct the reperfusion of middle cerebral artery occlusion in rats,to imitate the model of ischemic stroke.After 3 hours of awakening in model rats,a neurological function score was performed to determine whether the rats have successfully established the model.The rats with successful modeling were treated with experimental drugs,with the followings conducted at the end of the course of treatment;1.The neurological behavior scores of rats in each group were recorded and the recovery degree of brain damage in rats was determined.2.The morphological changes of neuronal cells in hippocampus area of rats were observed by HE staining,and the effects of the drugs in each group on brain tissue in rats with cerebral ischemia reperfusion were studied.3.Enzyme-linked immunosorbent assay:The content change of relevant factors SOD and MDA related to oxidative stress response in rat serum was detected;and the content change of cytokines IL-10 and TNF-? related to inflammatory reaction was detected.Results:1.Effects of SZXDS on neurological function in rats:Through the neurological behavioral score,it showed that neurological behavioral score in SZXDS drug group was lower than that in model group(P<0.05).2.Effects of SZXDS on neuronal cells in the hippocampus area of rats with cerebral ischemia reperfusion;Through the observation of neurons in the hippocampus of rats by optical microscope,it showed that SZXDS could improve the degree of neuronal cell injury,in which the cell interstitial edema of the neurons in the hippocampus area of the rat brain tissue in SZXDS high dose group was relatively reduced compared with that of the model,with the cell reduction in vacuolar structure and condensation.3.Effects of SZXDS on oxidability in rats with cerebral ischemia reperfusion:Compared with the model group,the content of SOD in rat serum of SZXDS high and low dose group increased(P<0.05),in which the content of SOD in rat serum of SZXDS high dose group was higher than that of SZXDS low dose group(P<0.05).Compared with the model group,the content of MDA in rat serum of SZXDS high and low dose group decreased(P<0.05),in which the content of MDA in rat serum of SZXDS high dose group was lower than that of SZXDS low dose group(P<0.05).4.Effect of SZXDS on inflammatory response in rats with cerebral ischemia-reperfusion:Compared with the model group,the content of IL-10 in rat serum of SZXDS high and low dose group increased(P<0.05),in which the IL-10 in rat serum of SZXDS high dose group was higher than that of the SZXDS low dose group(P<0.05).Compared with the model group,the content of TNF-a in rat serum of SZXDS high and low dose group decreased(P<0.05),and the content of TNF-a in rat serum of SZXDS high dose group was lower than that of SZXDS low dose group(P<0.05).Conclusion:SZXDS can play a protective role on brain tissue in rats with cerebral ischemia reperfusion,and its mechanism may be related to the enhancement of anti-oxidability of rats and the reduction of inflammatory reactions.
