BackgroundPituitary adenoma is a benign tumor with an increasing incidence in recent years,which is second only to glioma and meningioma in brain tumors About 40%of the pituitary adenomas in the clinic are invasive,because they often infringe the surrounding tissue and are difficult to remove completely in the operation,which is called invasive pituitary adenoma.In recent years,molecular therapy of invasive pituitary adenoma has become a new hot topic.Autophagy is discovered in recent years,a kind of different from apoptosis and necrosis,apoptosis,studies have shown that autophagy generally exists in a state of normal and pathological cell,autophagy and neurodegenerative diseases,immune cells and tumor cells of development there are inseparable relationship.In autophagy and the relationship of tumor and the role of autophagy and apoptosis is not clear that autophagy can through to their waste removal and provide nutrition function of inhibiting cell apoptosis,such as curcumin in the induction of laryngeal cancer Hep 2 cells apoptosis induced during the process of the protective autophagy occurs,at the same time autophagy can jointly participate in the process of cell death and apoptosis,such as in the process of cisplatin induced bladder cancer cell apoptosis autophagy can promote the apoptosis induced by cisplatin.Now reported in the literature in the specific drug under the action of some tumor cells had autophagy and the relationship between autophagy and the development of tumor cells,but whether autophagy in pituitary adenoma occurred and its relationship with tumor cells have not been reported.This test for wu benzene beauty effect of drugs,to verify the autophagy in pituitary adenomas of GH3 cells,and studied the autophagy and the specific role relationship between the tumor cells and tumor cells changes of molecular biology,molecular biology of pituitary adenoma treatment provides a theoretical basis.Objective to study the function and mechanism of autophagy in the process of the death of pituitary adenoma GH3 cells induced by ubenimex.Methods Cck-8 assay was used to determine the proliferation activity of cells after ubenemex at different concentrations,flow cytometry(FCM)was used to detect the apoptosis rate of cells in each group after ubenemex alone or in combination,LDH release test was used to determine cytotoxicity,rt-pcr was used to detect the mRNA expression of Beclinl and LC3,and Westernblot was used to detect the expression levels of lc-3,Beclinl,bcl-2 and p-akt proteins in GH3 cells.Results Cck-8 assay showed that ubenemex inhibited GH3 cell proliferation in a dose-dependent and time-dependent manner.Flow cytometry showed that ubenemex promoted GH3 cell apoptosis,which was reduced after the combination of autophagy inhibitor 3-ma.The results of LDH cytotoxicity test showed that with the increase of ubenemex,the cytotoxicity increased,and the cytotoxicity was reversed after the addition of 3-ma.Rt-pcr results showed that Beclinl and LC3 mRNA levels were significantly increased after ubamex treatment.Westernblot tests revealed wu LC-benzene beauty department significantly affect the cells,Beclinl,Bcl-2,and 3 p-Akt protein expression level,the Bcl-2 protein expression decreased,Beclinl protein expression,p-Akt protein expression decreased,LC3 II protein expression increased(p<0.05).Conclusions Ubenimex can induce the death process of Autophagic cell And the autophagy plays a role in promoting apoptosis,Beclinl?Bcl-2 and p-Akt can play a regulated role in the death process of pituitary adenoma GH3.
