Objective:To investigate the effects of transforming growth factor-?3(TGF-?3)and bone morphogenetic protein-2(BMP-2)on osteogenic differentiation of dental pulp stem cells(DPSCs).Methods:The pulp tissues from 4 healthy New Zealand rabbits were isolated and cultured by enzymolysis tissue mass method to obtain DPSCs.The morphology and growth of DPSCs were observed under microscope.The third passage DPSCs was divided into BMP-2 group and TGF-?3 group,added with 80 ug/L BMP-2and TGF-?3 culture solution,respectively.The expressions of collagen-?(COL-?)on the3~rdd and 7~thh days of culture and osteocalcin(OCN)in osteoblasts on the 7~thh and 14~thh days of culture were detected by immunohistochemical method.The integrated optical density(IOD)value was measured by Image-Pro plus(IPP)image analysis software.After induction of 7 and 14days,ALP activity was observed by inverted microscope on the 7~thh and 14~thh days after ALP staining in BMP-2 group and TGF-?3 group.Results:After 48 h of isolation and culture,the cells swam from the tissue block and the cells grew well.Most of cultures DPSCs were in fusiform or polygonal.After 3 days of induction,IOD value of COL-?was significantly higher in TGF-?3 group(159 676.799±63 959.568)than that in BMP-2 group(44 047.330±15 853.656)(P<0.05).There was no significant difference in the expression of IOC between TGF-?3 group(245 378.263±60 084.607)and BMP-2 group(223 294.430±80 266.723)on the 7~thh day of induction(P>0.05).The IOD value of OC was significantly higher in TGF-?3 group(109 568.817±14 079.885)than that in BMP-2 group(55 168.568±1 694.930)on the 7~thh day of induction(P<0.05),and there was no significant difference between TGF-?3 group(411 544.957±82 803.243)and BMP-2 group(360 103.540±20 835.362)on the 14~thh day of induction(P>0.05).On the 7~thh day of induction,a small amount of light bule ALP positive areas appeared in both groups,which increased in area and the blue color deepening on the 14~thh day of induction.There was no significant difference in ALP staining two groups.Conclusion:Compared with BMP-2,TGF-?3 is more effective to promote the osteogenic differentiation of rabbit DPSCs in early stage.
