Objective:The rat model of venous thrombosis of lower extremities was established through venous ligation surgery,and to compare the expression of IL-1?,NF-KB,XOD and TF in blood samples and model tissues,and to analyze the relationship between the above four factores and DVT.To analyze the mechanism of endothelial injury and thrombosis induced by EL-1?.Methods:2h,8h.24 h,48 h,72 h model group,sham operation group and normal group.2.the morphology of tissue and thrombus were observed by HE staining.3.the levels of IL-1?,XOD and TF in semm was detected by ELISA,and Flow cytometry was used to count CECs.4.the RT-qPCR was used to detect the relative expression of IL-1B,NF-kB,TF and XOD genes in the model segment tissue,and the Western Blot was used to detect the expression of NF-kB protein in model segment tissue.Results:1.all modeles was successfully established.2.results of HE staining in model group:tlirombosis observed in 2 h after operation.Fibroblasts were found in the thrombus 8 h after operation.A small amount of granulation tissue was observed at 24 h and 48 h after operation.72 h after operation,there was a large amount of granulation tissues.3.the levels of IL-IB,TF and XOD protein in serum,but model group was different from normal group and sham operation group(P<0.05).The concentration of CD31 in the model group was different from the normal group and shan operation group(P<0.05).4.the relative expression of IL-1?,TF,XOD and NF-KB genes,but model group was different from the nonnal group and sham operation group(P<0.01).There was no difference in the expression of NF-KB protein between nonnal group and sham operation group(P>0.05),but model group was different from normal group and sham operation group(P<0.05).Conclusion:1.IL-1B.NF-KB,TF,XOD are involved in the occurrence of DVT in rats.2.inflammatory factor IL-1?,by activating NF-KB,promotes the expression of XOD,Produce substances that damages endothelial cell,than exposed subendothelial collagen under the endothelial cell which activates TF and coagulation,and leads to thrombosis.
