Objective: To observe the effect of zhuang medicine ba du sheng ji ointment on wound healing in rats with refractory oxidative stress,and to explore its possible mechanism.Methods: 72 rats were randomly divided into control group,model group,Mr Complex economic positive control group,and poison paste the experimental group,the production of composite skin defect with epirubicin oxidative stress difficult wound in rats model was established,bandage daily after building,respectively,in 3 days,6 days,12 days calculated each rat wound healing rate,and serum and the newborn granulation tissue,to detect the contents of serum lysozyme by turbidimetric method,radioimmunity method to detect serum epidermal growth factor(EGF)and vascular endothelial growth factor(VEGF)levels,enzyme-linked immunoassay(ELISA)to detect the contents of EGF in newborn granulation tissue,Immunohistochemical method was used to detect the content of VEGF in the new granulation tissue.The pathological changes of the new granulation tissue after HE staining were observed under the light microscope,and the expression levels of PI3 K m RNA and AKT m RNA in the new granulation tissue were detected by fluorescence quantitative PCR.Results:1.Compared with the control group,model group wound healing rate,serum EGF and VEGF and lysozyme content,organization of EGF and VEGF levels,organizational PI3 Km RNA and AKTm RNA expression level,in addition to the three days after lysozyme and six days after tissue EGF content differences did not see statistical significance(P > 0.05),the rest were significantly less than control group(P < 0.05),the difference was statistically significant,suggesting making oxidative stress is difficult to heal wound model rats more successful.2.After intervention with beifuji and ba du sheng ji the wound healing rate increased significantly(P<0.05)after 3 days,6 days and 12 days,indicating that both beifuji and ba du sheng ji were effective therapeutic drugs.3.The detection results of serum lysozyme content showed that compared with the model group,the lysozyme levels in the beifuji group and the ba du sheng ji ointment group increased significantly after 3 days,6days and 12 days(P<0.05),with statistically significant differences.4.EGF and VEGF in serum and tissue of the results showed that: compared with model group,Mr Complex economy can improve serum 3,12 days and group6,12 days EGF and VEGF levels,pull out poison born ointment can improve serum after 3,6 days and group 6,12 days EGF and VEGF levels,the difference had statistical significance(P < 0.05),and suggest the complex economic and pull out poison born ointment can effectively improve the body EGF and VEGF levels,so as to promote wound healing.5.Histopathological results showed that: compared with model group,Mr Complex economic and toxic raw muscle pull cream group showing a rapid wound tissue edema disappeared,inflammatory reaction and rapid loss,granulation tissue growth faster,intervention 12 days newborn granulation tissue began to mature,some blood capillary occlusion,small artery,vein began to form,fibroblasts increased obviously,there are rules.6.The expressions of PI3 Km RNA and AKT m RNA in the new granulation tissue showed that,compared with the model group,the expressions of PI3 Km RNA in the beifuji group were significantly increased after 3 days,6 days and 12 days,and those in the beifuji group were significantly increased after 6 days(P<0.05),and the differences were statistically significant.Conclusion: 1.Zhuang medicine ba du sheng ji ointment can promote wound healing in rats with refractory oxidative stress.2.The zhuang medicine ba du sheng ji ointment can increase the contents of EGF,VEGF and lysozyme in the serum of rats with refractory oxidative stress wounds.3.The zhuang medicine ba du sheng ji ointment can increase the content of EGF and VEGF in the tissues of rats with refractory oxidative stress wounds.4.Ba du sheng ji ointment,an zhuang medicine,can improve the growth of new granulation tissue in rats with refractory wounds caused by oxidative stress.5.Zhuang medicine ba du sheng ji ointment may promote wound healing by activating the PI3K/AKT signaling pathway.
