Objective: Through the observation on the zhuang medicine Longzuantongbi Decoction on osteoclast apoptosis rate and quantity of TLR4,Myd88 protein expression and TNF-?,NFAT-c1,c-Fos gene expression,the influence of understanding patterns of migration,differentiation and apoptosis of osteoclasts,will be able to better prevention and treatment of rheumatoid arthritis bone destruction,discusses zhuang medicine Longzuantongbi Decoction through the side of the mechanism of action of rheumatoid arthritis.Methods:In vitro aseptic separation of C57BL/6 mice femoral bone marrow mononuclear cells,m-csf and RANKL combined culture induced osteoclast,adding drug-containing serum intervention,divided into the normal group,western medicine group,zhuang medicine Longzuantongbi Decoction prescription high dose,medium dose,low dose group.Tartrate resistant acid phosphatase(TRAP)staining to specific morphological staining of osteoclasts,using flow cytometry to detect each group containing medicine serum of osteoclast apoptosis rate,the influence of observation on the zhuang medicine Longzuantongbi Decoction through polioplus partners party osteoclast apoptosis rate,and using Western Blot method to detect osteoclast TLR4?Myd88 protein expression in quantity,using reverse transcription polymerase chain reaction detection of osteoclast TNF-??NFAT-c1?c-Fos gene expression.Results: 1.Results of osteoclast apoptosis rate: compared with the normal group,the apoptosis rates of the western medicine group,the zhuang medicine Longzuantongbi Decoction prescription group and the middle dose group were all increased,with statistically significant differences(P<0.05);Compared with the normal group,there was no difference in the low dose group of zhuang medicine Longzuantongbi Decoction prescription,and the difference was not statistically significant(P>0.05).2.Expression levels of TLR4 and Myd88 proteins in osteoclasts: compared with the normal group,the expression levels of TLR4 proteins in the western medicine group,the zhuang medicine Longzuantongbi Decoction prescription high dose group and the middle dose group were all decreased,with statistically significant differences(P<0.01);Compared with the low dose group,there was no significant difference between the normal group and the low dose group.3.TNF-? in osteoclast,NFAT-c1,c-Fos gene expression level: compared with normal group,western medicine group,the zhuang medicine Longzuantongbi Decoction through certification of the high dose group of TNF-?,NFAT-c1,c-Fos gene expression were cut,statistically significant difference(P<0.01);the zhuang medicine Longzuantongbi Decoction through polioplus partners party dose group of TNF-?,NFAT-c1,c-Fos gene expression,statistically significant difference(P<0.05)and normal group and the zhuang medicine dragon drill through bi low dose group was no significant difference,the difference was not statistically significant.Conclusion: 1.Zhuang medicine Longzuantongbi Decoction can increase the rate of osteoclast apoptosis and reduce the activity of osteoclast,which can better treat and prevent the bone destruction of RA;2.Zhuang medicine Longzuantongbi Decoction can reduce the expression levels of TLR4 and Myd88 proteins in osteoclasts,and block the TLR4/Myd88 signal transduction network,resulting in decreased osteoclast activity;3.Zhuang medicine Longzuantongbi Decoction downregulates the expression of TNF-?,NFAT-c1 and c-Fos genes in osteoclasts,thereby affecting the differentiation of osteoclasts.
