As a new high-performance nanomaterial,Zinc oxide nanoparticles(ZnO NPs)are used in an increasing number of industrial products such as paint,coating,cosmetics,food,pharmaceutical products and other applications.There have been many suggestions of a ZnO NPs toxicity paradigm but the underlying molecular mechanisms about the toxicity of ZnO NPs remain unclear.This study was done to find the potential biological functions of ZnO NPs and to provide new technology and perspectives for the development of modern biology and medicine.Firstly,we studied the autophagy induced by ZnO NPs in HeLa cells.In this paper,we found that ZnO NPs can improve the level of autophagy by a systematic experiments such as static and dynamic state experiments assay:by detecting fluorescence dot accumulation in EGFP-LC3/HeLa cell and the rate of LC3-II in Hela cell,both of which increased in a concentration-and time-dependent manner after ZnO NPs treatment,together with the result caused by autophagy inhibitor(CQ and wortmannin),we demonstrated that ZnO NPs induced a full autophagic flux.preliminary ZnO NPs may cause lysosome biogenesis and increase the expression of lysosomal hydrolases Cathepesin D both in mature and non-mature form,Accordingly,it is also through autophagy pathway.Then we studied and discussed sensitized chemotherapeutic killing effect of ZnO NPs in the treatment of cancer chemotherapy.Further study show that ZnO NPs led to cell death through Dox in Hela cells was related to autophagy induction,as determined by cell death and Western Blot along with autophagy inhibitor,the results highlight and present the application of ZnO NPs.providing a new perspective of ZnO NPs in cancer therapy.
