Objective: To explore the effects of IL-17 to promote proliferation,differentiation and apoptosis of RAW264.7.Methods: RAW264.7 cells were cultured by RANKL and M-CSF at 24 h after treated with IL-17.CCK-8 kits and RT-PCR were used to evaluate the effects of proliferation and differentiation,Flow cytometry were used to evaluate the effects for apoptosis.Results: IL-17 could promote proliferation of RAW264.7 cells,the proliferation effects of IL-17 had a dose-effect relationship and was in a time-dependent manner;At the concentration of 50ng/mL of IL-17,the activity of genes including Cath-K,RANK and NFATC1 mRNA increased,inhibition of apoptosis.Conclusion: It is demonstrated that IL-17 mainly promotes RAW264.7 cells proliferation and inhibition of apoptosis,and promotes that RAW264.7 cells differentiate into osteoclasts and plays a key role in bone-remolding. |
