Objective: To investigate the expression of Annexin A3(ANXA3)in osteosarcoma cell line HOS by Celastrol,to explore the mechanism of inhibition on osteosarcoma cells,and to find a new method for immune-targeted therapy of osteosarcoma.Methods : Western blot was used to detect the expression of ANXA3 in osteoblasts and HOS?U2OS cells at first.Then Celastrol was used to culture osteosarcoma HOS cells.The morphology of each groups were observed by phase contrast microscopy,and the cell viability was detected by Cell Counting Kit-8(CCK-8)assay.The expression levels of ANXA3 and LC3 B proteins in each group were detected by Western blotting.Finally,the apoptosis rate of osteosarcoma cell line HOS was performed by flow cytometry.Results:The expression of ANXA3 in osteoblasts,osteosarcoma cell lines HOS and U2 OS were confirmed by Western blotting,the expression of ANXA3 in HOS and U2 OS cells were significantly increased,especially in HOS cells.Celastrol changed the morphology of osteosarcoma cell line HOS,inhibited the proliferation in a dose-dependent manner.In addition,it could promote their apoptosis,activate autophagy-specific protein LC3 B,and inhibited theexpression of ANXA3.Conclusions: Celastrol activated LC3 B expression by inhibiting ANXA3 and induced apoptosis of osteosarcoma cell line HOS.High dose of Celastrol can effectively kill osteosarcoma HOS cells.
