Circulating tumor cells are all kinds of tumor cells existing in peripheral blood.They are separated from solid tumor lesions by spontaneous or diagnostic procedures.Detection of circulating tumor cells plays an important role in the treatment and prevention of cancer.Although a variety of circulating tumor cell detection methods and products have been designed,most of these methods need to go through many complex steps,which may lead to the destruction of tumor cells in the process of detection,affecting the accuracy of detection results.Therefore,in this paper,we merged the dark field imaging device with the circulating tumor cell recognition,and developed an experimental device to detect circulating tumor cells by scanning imaging,to identify circulating tumor cells in patients'peripheral blood.Referring to the principle of CD disk,the scanning spot rotates while the sample surface is shifted.It can scan the surface of the biological sample with large area and high resolution in a short time.While ensuring accuracy,the efficiency of detection is greatly improved.However,due to the influence of the large scanning beam and the surface transmittance of biological samples,the traditional scanning imaging experimental light path can not accurately scan the signal of circulating tumor cells.Therefore,in order to improve the contrast and resolution of scanning,we have improved the optical path,adding a small aperture and ellipsoid mirror in the optical path,thus forming an ellipsoid confocal dark field scanning experimental optical path.In the confocal light path of ellipsoidal mirror,the scattered light signal of the ellipsoidal mirror can be obtained by placing the working spot in one focal point of the ellipsoid.By collecting and processing this signal,the cell distribution on the surface of the circulating tumor cell sample can be obtained.After the scanning,we automatically recognize the image according to the intensity of the scanning signal and the size of the imaging spot,and select biological samples with circulating tumor cells from a large number of peripheral blood samples.By using this method,the peripheral blood samples to be detected need not complex biological enrichment process,reducing the damage and destruction of the circulating tumor cells,thus improving the detection accuracy and further improving the accuracy of the scanning results.
