Abstract/Summary:
Objectives:As edible fungi,Dictyophora indusiata has high edible value and healthy function,which is relatively short of resources.Study to the fruiting body can be replaced by using solid fermentation method to cultivate mycelium of Dictyophora indusiata to study its secondary metabolites and active substances.The anti-inflammatory part from mycelium of Dictyophora indusiata will be further traced,screening out the small molecular compound with anti-inflammatory activity,which can be provide the basis for the quality assessment of Dictyophora indusiata,and laying the foundation for the discovery of new natural anti-cancer drugs or lead compound of food preservative.Methods:Using solid fermentation method to cultivate Dictyophora indusiata strains,and fermentation products were extracted with ethyl acetate’s soak,getting thick paste by reduced pressure,adding water to disperse,Using petroleum ether,ethyl acetate,n-butanol respectively to extract,recovering by reduced pressure,separating it through silicagel column,gel column,ODS-column,the monomeric compound was obtained,analyzing it with 1H-NMR and 13C-NMR atlas.In the experiment of immunocompetence,choosed Raw264.7 megalophage and HepG2 hepatoma cell respectively to study the immunocompetence and anti-tumor of Dictyophora indusiata.Results:Five compounds were isolated and identified,which wereβ-sitosterol,Methyl Oleate,Alternariol monomethyl rther,stigmasterol,palmitic acid.The cell experiment showed that petroleum ether fraction had growth inhibitory impact to HepG2 hepatoma cell.The Inhibition maximum was 73.08%in 200μg/mL,72h.Ethyl acetate fraction had obvious immunological enhancement to Raw264.7 mouse peritoneal macrophage.Thecell immunocompetence of five monomeric compounds were measured in turn.Alternariol monomethyl rther and Methyl Oleate had different immunocompetence level HepG2 hepatoma cell.Conclusion:The cell experiment showed that ethyl acetate of solid fermentation products from Dictyophora indusiata was the main fraction to play anti-inflammatory and anti-cancer activity.Through the active verification of monomeric compound,small molecule was the main element to play immunization.Alternariol monomethyl rther and Methyl Oleate were also traced as important monomeric compounds.This laid the foundation for the quality evaluation of Dictyophora indusiata and new drug development.
