Objective:To study the dynamic changes of major hypoglycemic active ingredients in the process of germination of tartary buckwheat with trace elements,and to study the preliminary pharmacodynamics of the tartary buckwheat hypoglycemic activity after germination,provide a theoretical basis for the development of new types of hypoglycemic resources.Method:Using inositol content as an indicator,the best method for extracting inositol from tartary buckwheat was obtained by comparison of water extraction and ethanol extraction,through the pure water germination,germination of trace elements,acid hydrolysis,alkaline hydrolysis and other methods to get the best way in the inositol derivatives of tartary buckwheat,the germination conditions of trace elements were optimized by response surface methodology,an HPLC-ELSD was used to monitor changes in inositol content during germination and plot the change curve.The content of flavonoids in the germination process was monitored by HPLC with the content of rutin and quercetin as indicators.Hydrochloric acid-n-butanol method was used to monitor the changes of procyanidins in the process of buckwheat germination;Inductively Coupled Plasma Mass Spectrometry?ICP-MS?was used to determine the content of inorganic trace elements and organic trace elements after the addition of trace elements in tartary buckwheat and the organic conversion of inorganic trace elements;Alloxan-induced diabetic mice as a model,the mouse blood glucose as an indicator to study the effect of tartaric and malondialdehyde-induced diabetic mice hypoglycemic effect before and after the micro-element germination.Results:The best method for extracting bitter quinonesitol is alcohol extraction.The best extraction process is to use 40 times 40% ethanol solution at 85?for 3 hours.The optimal conditions for the germination of trace elements were as follows: the mixed solution of chromium trichloride with mass concentration of 80mg·L-1,manganese sulfate with 500mg·L-1,and sodium selenite with concentration of 30mg·L-1 was the soaking solution and spray liquid,soaking time 6h,germination temperature 25?,germination time is 4 days.Inositol extraction of the best process for the 40 times the amount of 40% ethanol at 85? under reflux extraction 3h,during the process of buckwheat germination,the content of free inositol gradually increased and then decreased,reaching the maximum at the fourth day of germination;the content of flavonoids increased gradually and then decreased in the process of buckwheat germination.,the total amount of rutin and quercetin reached the maximum on the 4th day.Proanthocyanidins content in the germination process was gradually increased and then decreased,reaching the maximum on the 4th day.On the 4th day of germination,buckwheat contained organic chromium 76.53?g·g-1,organic manganese 265.2?g·g-1,and organic selenium 32.70?g·g-1;the addition of trace elements in buckwheat can significantly reduce fasting blood glucose levels in diabetic mice after germination.Conclusion:The germination of trace elements had the best transformation effect on tartary buckwheat myo-inositol derivatives,and the inositol content after transformation increased by 60 times of the original concentration,the content of rutin increased,the content of quercetin increased first and then decreased,the content of total anthocyanin showed an increasing trend,significant increase in organic trace elements after the addition of trace elements,and the content of quercetin had significant hypoglycemic activity after the transformation.
