Objective: To observe the therapeutic effect of acupuncture on amblyopia in children and investigate the intervening effect of acupuncture on the model of monocular deprivation amblyopia rats and to explore the different patterns of changes between actin dynamics and synaptic density in the rat’s primary visual cortex during a special period of visual development.Methods: The study included three experiments.Experiment 1: Therapeutic effect of acupuncture on children with amblyopia.To study the effect of acupuncture on the treatment of children with amblyopia,we selected 81 children from May 2016 to May 2017 to our hospital for treatment of amblyopia.They were divided into the control group and the treatment group.Experiment 2: The effect of acupuncture on the model of monocular deprivation amblyopia rats.The experimental animals were healthy SD rats at 13 days(P13),30 days(P30)and 180days(P180)after birth.The monocular deprivation amblyopia modes were established at P13.After successful,they were randomly divided into amblyopia group and acupuncture group(15 animals in each group);acupuncture intervention were began at P30 on acupuncture group;when acupuncture intervention were completed,both of amblyopia group and acupuncture group began to behavioral vision test.Experiment 3: The different patterns of changes between actin dynamics and synaptic density in the rat’s primary visual cortex during a special period of visual development.The experimental animals were healthy SD rats and were divided into P15 group,P30 group and P45 day(18 animals in each group).The change of synaptic density in V1 B was detected by transmission electron microscopy.The expression of PSD-95 proteins in V1 was detected by western blot.Results:1.The total effective rate of the treatment group was 92.3% and 72.2% in the control group.The difference between the two groups was statistically significant(X2 = 150.000,P = 0.000).The treatment effect of the treatment group was significantly better than the control group(u = 3.097,P = 0.000).2.Visual acuity of acupuncture group was significantly higher than that of amblyopia group(t =-5.227,P = 0.00).3.There was no significant difference in the synaptic density of V1 B between P30 and P45(t = 0.477,P> 0.05).There was no significant difference in the expression of PSD-95 protein of V1 between P30 group and P45 group(one-way ANOVA: F(2,6)= 7.606,P <0.05;P30 vs.P15,P <0.05;P45 vs.P15,P <0.05;P45vs.P30,P> 0.05).Conclusion:1.On the treatment of amblyopia,the role of acupuncture can not be ignored;acupuncture combined with other therapies should be the ideal treatment for amblyopia.2.This device can greatly reduce the influence of the restraint itself during the experiment and can be popularized in the acupuncture experiments;for the first time the method of animal behavior vision test applied to acupuncture treatment of deprivation amblyopia rats.It lays a solid foundation for the research of molecular experiments in the future.3.During visual development the changing patterns between actin rearrangement and the synaptic densities are interrelated but not absolutely identical.Actin rearrangement reflects both functional and structural synaptic plasticity.In addition,the synaptic pruning process may occur and play more roles in the development of eye-specific retinal connections.
