As a multi-ethnic country,China is rich in national medicines,which are the indispensable treasure of Chinese Traditional Medicine.National medicine has been used for thousands of years and its clinical efficacy is clear and reliable.However,the basic research of ethnic medicine is far less than that of traditional Chinese medicine,limiting the development and appliance.Therefore,this study studied the chemical composition and biological activity of two ethnic medicinal plants,Uigur Medicine Morus nigra and Miao Medicine Laportea bulbifera,to provide a certain scientific foundation for the further development and use of them.This study is made up of three parts:the research progress of chemical constituents and biological activities of uigur medicine Morus nigra and Miao Medicine Laportea bulbifera;the study of chemical constituents and biological activity of Uigur Medicine Morus nigra;the study of chemical composition and anti-inflammatory activity of Miao Medicine Laportea bulbifera(Sieb.Et Zucc.)Wedd..Chapter 1:Research Progress of Chemical Constituents and Biological Activities of Uigur Medicine Morus nigra and Miao Medicine Laportea bulbifera Morus nigra,as an important medicinal plant of Morus genus,mainly grows in Xinjiang.It is also the only 22-ploid species in the genus Morus.Its main chemical components consist of prenyl flavonoids,terpenoids,polyphenols,coumarins,steroids,and alkaloids,and its major biological activities consist of hypolipidemic,hypotensive,anti-oxidative,anti-diabetic,anti-inflammatory,tyrosinase inhibitory,and anti-virus activities.Laportea bulbifera(Sieb.Et Zucc.Wedd.),belonging to the genus Laportea,is used as root or the whole plant.It contains multiple chemical components,including coumarin,flavonoids,tannins,steroids,oils,and fats,which exhibite analgesic,anti-inflammatory,immunosuppressive,antioxidant,and anti-diabetic effects.Chapter 2:Study on Chemical Composition and Biological Activity of Uigur Medicine Morus nigra1.Study on Chemical Composition of Morus nigraThe pre-experiment indicated that the dichloromethane portion of M.nigra extract showed prominent inhibitory effect on a-glucosidase,while the ethyl acetate portion had better tyrosinase inhibitory activity.Therefore,in order to clarify the substance basis of the bioactivity and seek for new active compounds,these two portions were studied for the chemical composition.Separation methods,including macroporous resin column,silica gel column,reversed-phase C18 silica gel column,MCI column,Sephadex LH20 gel column,and semi-preparative HPLC,and spectral analysis means,including IR OR,MS,NMR and CD,were used to separate and identify compounds from M.nigra.The study yielded fifty-three compounds,including flavonoids,terpenoids,DA adducts,polyphenols,steroids,esters,and alkylenes.Among them,twelve compounds(mn-1-mn-10)were determined as new natural products.mn-1 was speculated as the biogenetic precursor,and the biogenetic pathway from mn-1 to sanggennol F was speculated in this study.mn-5-a and mn-5-b were the skeleton unit structure of flavonoid flavonoids.mn-2,mn-3,and mn-9,mn-10 were the 4th pair and the 5th pair of sanggennone type flavonoid epimers isolated from M.nigra.mn-6 was the first benzofuranone structure with isopentenyl,which was also the first compound isolated from Morus plants.mn-8 was a flavonol structure with a highly cyclized structural unit which was very rare in flavonoids and has been prepared by chiral resolution to give a pair of enantiomers naming mn-8-1 and mn-8-2.2.Study on Biological Activity of Morus nigra?PPAR ? agonistic activityFour compounds(mn-2,mn-4,mn-14,mn-18)with PPAR y agonist activities were screened out by dual luciferase assay.The structure-activity relationships of related compounds were summarized.Further experiment showed that the new compound mn-2 was a potential PPAR ? partial agonist.??-glucosidase inhibitory activity Seventeen a-glucosidase inhibitors were screened out from M.nigra by spectrophotometry.On this basis,the structure-activity relationships of flavanone-type flavonoids were investigated.?Tyrosinase inhibitory activity The ethyl acetate fraction of M.nigra,identified as tyrosinase inhibitor during the pre-experiment,was further investigated in cellular experiment The result showed that it had a trong inhibitory effect on the secretion of melanin from mouse melanoma B16 cells.Moreover,some active compounds were identified with the help of HPLC fingerprinting.Chapter 3:Study on Chemical Composition and Anti-Inflammatory Activity of Miao Medicine Laportea bulbifera(Sieb.Et Zucc.)Wedd.Miao medicine Laportea bulbifera(Sieb.Et Zucc.)Wedd,belonging to the Ramie family,is mainly produced in Guizhou,and its traditional medicinal use consists of blood circulation and hurricane itching.It is used to treat rheumatism and pruritus in folk.1.Study on Chemical Composition of Laportea bulbifera(Sieb.Et Zucc.)Wedd Seven compounds,including four steroids and three long-chain fatty acids,were separated from Laportea bulbifera(Sieb.Et Zucc.)Wedd by multiple separatal and identifical methonds mentioned above.All of the compounds were firstly isolated from the genus.2.Study on Anti-Inflammatory Activity of Laportea bulbifera(Sieb.Et Zucc.)Wedd All of the four steroids showed certain anti-inflammatory activities in cell model evaluation,out of which,Lb-1 were the best candidate.In this study,the research on the chemical composition and related activities of M.nigra and Laportea bulbifera supplemented and perfected the basic research of the two medicinal plants.And it clarified the material basis of their efficacy and provide a certain theoretical basis for further development and study.
