Objective :To study the level and significance of T helper 17(Thl7)CD4+CD25+ Foxp3+ regulatory T cells(Treg)and miR-155 in peripheral blood of Acinetobacter baumannii sepsis rats and explore their relationships?Methods :The Acinetobacter baumannii rat model was established by intraperitoneal injection with Acinetobacter baumannii.Separation of rat blood lymphocytes,fluorescence quantitative rt-pcr was used to detect expression of miR-155.Flow cytometry was used to detect the ratios of peripheral and Treg and Th17 cells over CD4+T cells.Peripheral interleukin-17(IL-17)and Peripheral interleukin-10(IL-10)was measured by enzyme-linked immunosorbent assay(ELISA).TGF-?was measured by enzyme-linked immunosorbent assay(ELISA)?The differences were compared between groups of 6h,12 h of sepsis,24 h of sepsis and control group.Pearsons correlation analysis was used to evaluate the relationship between miR-155?Treg?Th17?IL-17?IL-10 and TGF-?.Results:1.Compared with the normal control group,the miR-155 level of the rats with sepsis was elevated(all P<0.01)and reached the peak at 12 h.2.Compared with the normal control group,the levels of TH17 and Treg cells in rats with sepsis were higher(all P<0.01)and reached the peak at 12 h.3.Compared with normal control group each time sepsis rats IL-17,IL-10,TGF-? levels,12 h IL-17 in plasma peak,to observe the expression of IL-17 from the 12 h to 24 h expression in plasma volume flatten out.The expression of TGF-? in plasma was rapidly increased in 6h,and TGF-? in the time period of 6h to 24 h was gradually reduced.The expression level of IL-10 in plasma reached a peak in 12 h,and the expression of IL-10 in plasma was decreased from 12 h to 24 h.4.Pearson correlation analysis showed that the expression of miR-155 was positively correlated with the ratio of Th17,Treg cells(P<0.01).Pearson correlation analysis showed that the expression of miR-155 was positively correlated with IL-17,IL-10 and TGF-? level(P<0.01).Conclusions : The expression of Th17 ? Treg and miR-155 was increased in Acinetobacter baumannii sepsis rats.The expression of IL-17?IL-10 ?TGF-?was increased in Acinetobacter baumannii sepsis rats.serum level of miR-155 was positively correlated with Treg and Th17 cells and their related cell factors which can used as the target in the early diagnosis and treatment of Acinetobacter baumanni. |
