Objective:To study the differential expressions of Aldehyde dehydrogenase 2 gene and lipid metabolism related genes between leprosy patients and healthy controls.To study the expression of ALDH2 gene in mouse macrophages stimulated by Mycobacterium marinum in vitro by cytology experiments,and to investigate the correlation between ALDH2 gene and leprosy.Methods Select tissue from 27 leprosy patients lesions and 18 healthy controls skin,and the expressions of lipid metabolism related genes mRNA from them were compared by comparative RN A sequencing.The expression of gene ALDH2 mRNA levels in the peripheral blood of 18 leprosy patients and 26 healthy controls were determined using quantitative PCR.And concentrations of ALDH2 in the serum of 27 patients and 18 controls were measured by enzyme-linked immunosorbent assay.Stimulate the macrophage of mouse making use of different concentrations of fish mycobacteria,then RNA was extracted from infected cells and transcript to cDNA,and finally,the expressions of ALDH2 gene at different times were determined with method of qPCR.Results The expressions of 22 selected genes involved in the lipid metabolism showed remarkable differences between patients and controls(P<0.05).The difference of ALDH2 mRNA levels in peripheral blood was not significant between leprosy patients and healthy controls(P = 0.50).Concentration of ALDH2 in leprosy patients' serum was obviously lower than that in healthy controls(P<0.01).Cytological experiments showed,as the amount of bacteria in the macrophages of mouse infected by fish mycobacteria increased,the quantity of ALDH2 increased.When the proportion(MOI)between bacteria mycobacterium and macrophages cells reached 50,the quantity of ALDH2 in mouse macrophages cells greatly increased,comparing with that without stimulation.Conclusion Lipid metabolism related genes were remarkably differentially expressed between leprosy patients and healthy controls.The concentration of ALDH2 was significantly lower in leprosy patients than that in healthy controls.The expression quantity of ALDH2 in the infected cells increased significantly.It is possible that ALDH2 affected the onset and development of leprosy by regulating lipid metabolism.
