Objective:To explore the influence of isoliquiritigenin(ISL)on the proliferation,apoptosis of human glioma U251 cells and its molecular mechanisms thereof,provide reliable theoretical basis for future clinical application.Methods:The logarithmic phase human glioma U251 cells were divided into control group(normal cell culture fluid)and isoliquiritigenin group(isoliquiritigenin soluble in cell culture fluid,the concentration respectively was 10,20,40,60 and 80?M),each group had five repeat holes.The proliferation of U251 cells were detected with MTT assay after 12h,24h and 48h of culture,groped for subsequent experimental drug concentration and action time;The apoptosis of U251 was assessed with flow cytometry and the invasion abilities of U251 cells was tested with Transwell test;The expression levels of apoptosis related protein Bcl-2 and Bax,invasion related proteinsas MMP-2 and MMP-9,as well as phosphatidylinositol 3-kinase/protein kinase B(PI3K/Akt)pathway protein Akt and P-Akt were determined by Western blot assay.Result:Compared with the control group,various concentrations of ISL inhibited U251 cells proliferation in a dose-dependent manner and time-dependent manner,the half inhibitory concentration(IC50)of12?24?48 h respectively was 58.93 ±5.14?39.46 ± 3.41.29.58 ± 2.83 p.M,so we chosed 20?40?60?M isoliquiritigenin as subsequent experimental drug concentration and 24h as action time.Compared with the control group(2.35±0.61)%,the apoptosis rate of 20 ?M(8.93±1.94)%.40?M(14.84±2.11)%.60 ?M(27.63± 3.43)%increased gradually(P<0.05).Compared with the control group(322.75±13.41),the membrane cell number of 20 ?M(224.50± 10.35).40?M(126.30±17.56)?60 ?M.(62.01 ±7.08)decreased gradually(P<0.05).There was no statistically significant difference between groups of the expression level of protein Akt;The expression level of P-Akt?MMP-2?MMP-9?Bcl-2 decreased gradually and Bax increased gradually With the increase of the isoliquiritigenin concentration(P<0.05).Conclusion:Appropriate concentration of isoliquiritigenin can obviously inhibit the proliferation and induce apoptosis of U251 cells,which may be related to the down regulation of the signal transduction pathway protein PI3K/Akt.
