Objective Breast cancer is a malignant tumor in the world of serious harm to women's health,in many developed countries has been ranked first in female malignant tumor death fat breast cancer,there are two aspects,one is the increase in the incidence,on the other hand it is easy to relapse.Indirubin,also known as the artillery artillery,has an inhibitory effect on the tumor cells.The aim of this study is to verify the inhibitory effect of indirubin on human breast cancer MCF-7 cell line at cellular level,and to explore whether it can reverse the drug resistance of drug-resistant cell line.Method The purchase of indirubin,in breast cancer cell MCF-7 as the research object,to determine the inhibitory effect of indirubin on the proliferation of MCF-7 and the optimal concentration at the cellular level;from the perspective of cell proliferation and apoptosis,preliminary study on the mechanism of growth inhibition of MCF-7 cells of indirubin.On the basis of above,the adriamycin resistant breast cancer cell line MCF-7/Adr was used to verify whether indirubin could increase the sensitivity of the cell to adriamycin,and detect the expression of drug resistance protein by Western blotting.Results The survival rate of MCF-7 cells was significantly decreased by adding 10?g/mL,50?g/mL and 100?g/mL,and the survival rate of MCF-7 decreased with the increase of indirubin concentration.The inhibitory effect of the same concentration of indirubin on MCF-7 cells was time dependent,and the inhibition effect was better when the time was prolonged.Compared with the control group,the number of apoptotic cells in the indirubin group increased significantly,the number of G0-G1 phase cells increased and the S cells in the split stage decreased significantly.When the concentration of doxorubicin is 50?g/mL,the combination of indirubin can play a secondary role.When the concentration of doxorubicin is 75 and 100?g/mL,the combination of indirubin can play a synergistic role.After the use of indirubin,the IC50=20.61 of MCF-7/Adr cells and the resistance index of =21.25 were significantly lower than that of 25.62 before the use of indigo red.After imposing indirubin,the expression of drug resistant protein P-gp in drug resistant cell MCF-7/Adr decreased significantly.Conclusion Indirubin can inhibit the growth of MCF-7 in human breast cancer cells by inhibiting proliferation or inducing apoptosis.Indirubin can inhibit the growth of adriamycin resistant human breast cancer cell MCF-7/Adr and reverse its drug resistance,which is realized by reducing the expression of P-gp resistant protein.
