Oyster is an important cultured shellfish in China,with high nutritional value and medicinal value.Oysters used to treat diabetes in many traditional Chinese medicine prescriptions and medical practice.Determining the oyster hypoglycemic medicinal site has a pivotal role for its development as a drug.Polysaccharides in oysters is an important active substance,it has good antioxidant activity.Antioxidant activity is often associated with hypoglycemic effect.Therefore,oyster polysaccharides with antioxidant activity have potential hypoglycemic effects.In this paper,oyster polysaccharides(CGPS)as the research object,the use of metabolomics method to study CGPS hypoglycemic mechanism.The safety toxicology of CGPS was also studied.In order to provide the basic data for the development of CGPS drug development.Specific work is as follows:1.Fifty mice successfully selected were randomly divided into hyperglycemia group and positive group.In the low,middle and high CGPS group,10 mice in each group and 10 mice in the other group were normal group.The administration group was administered twice a day.The normal control group and the model control group were given distilled water twice a day for 6 hours at intervals of 21 days and fasted for 12 hours after administration.Physiological and biochemical indexes of the mice at the 7th,14th and 21st days were measured.Compared with hyperglycemia group,the fasting blood glucose of mice in low,medium and high dose groups decreased,compared with normal group,TG and TC content of positive control group,low dose group and middle dose group did not change significantly,The results showed that oral administration of metformin hydrochloride,low dose or medium dose of CGPS can maintain normal levels of TG and TC in mice.The results showed that different dosages of CGPS could affect body weight,drinking water,food intake and blood sugar of mice in varying degrees.With the increase of CGPS,the comprehensive efficacy evaluation increased,and the comprehensive medicine of CGPS in high dosage group.Effective evaluation of up to 33.26%.2.Urine samples from control mice(XYC),model mice(XYM),and therapeutic mice(XYP)were isolated and collected using HILIC-Q-TOF MS.The model group compared with the blank group(XYM-XYC),positive and negative ions obtained 13 kinds of potential markers.The treatment group compared with the model group(XYP-XYM)group positive and negative ions obtained 19 kinds of potential markers.The increase in the amounts of L-carnitine and hippuric acid indicates that the mechanism by which CGPS has a therapeutic effect on diabetes is that it regulates the glucose metabolism of model mice and improves their intestinal microflora.3.In vitro mammalian TK gene mutation test results showed no CGPS on mammalian cell TK gene mutations in vitro.CGPS in SD rats 28d oral toxicity test,no CGPS on SD rats have a potential toxic effects,no CGPS on SD rats there is a clear target organ toxicity,CGPS on SD rats 28 An oral toxicity test is not harmful at a dose of 6.75 g/kg BW.The above results show that CGPS has the mechanism of action for the treatment of diabetes may be to regulate the model mice glucose metabolism and improve its intestinal flora.CGPS have good safety.Therefore,CGPS has the potential as a drug development and utilization of value.
