AimTo aim at exploring the function and mechanism of LncRNA BC200 in esophageal carcinoma(EC)cells,we analyze differentially expressed genes(DEGs)of EC cells with BC200 knock-down and take the bioinformatics analysis of DEGs.MethodsEC cell line EC9706 cells were divided into BC200 knock-down group,negative control group and blank control group.EC9706 cells in the BC200 knock-down group and negative control group were infected with lentivirus vectors with BC200 targeted interfered sequence and unrelated sequence,respectively.The infected rate and interfered efficiency were measured with a fluorescence microscope and real-time quantitative PCR(qRT-PCR),respectively.DEGs of EC9706 cells with BC200 knock-down were analyzed with DNA microarray technology.Moreover,we used IPA software to analyze canonical pathways,disease and functions,and gene interaction networks enriched with DEGs.And the screened genes with higher fold change(FC)in the gene interaction network of carcinoma were validated by qRT-PCR.ResultsIn the study,we found that lentivirus-mediated interference sequence with targeting BC200 and irrelevant sequence could successfully infect EC9706 cells,and the expression of BC200 in EC9706 cells of BC200 knockdown group was significantly lowered than the control groups(P<0.001).DEGs analysis of DNA microarray data revealed 406 DEGs with P<0.05 and |FC|>1.5 as screening criteria,comprising 91 upregulated genes and 315 downregulated genes.IPA showed that the top three canonical pathways were estrogen receptor(ER)signaling,PI3 K Signaling,and Thrombin Signaling.IPA revealed that the top three disease and functions were Cancer,Organismal Injury and Abnormalities,and Gastrointestinal Disease.IPA indicated that the most inhibited upstream regulator was ATF4(Z=-4.119),the most activated upstream regulator was TRIB3(Z=3.268).IPA showed that the top one gene interaction network mainly affected Cancer,Cell Morphology,and Cellular Function and Maintenance(Score=50).6 screened genes with higher |FC| in the gene interaction network of Cancer were validated by qRT-PCR and the result revealed that the expressed level of CHAC1 and STC2 in the BC200 knock-down group was significantly lowered(P<0.05),which was consistent with the analysis result of DNA microarray.Conclusion1.In EC9706 cells with BC200 knock-down,406 DEGs is screened by analysis of DNA microarray data,comprising 91 upregulated genes and 315 downregulated genes.2.BC200 is involved in various canonical pathways in EC9706 cells.The upstream regulators in these pathways,such as ATF4 and TRIB3,may be the target of diagnosis and treatment of esophageal carcinoma.3.BC200 may play a role in EC9706 cells by up-regulating CHAC1 and STC2 genes in the ER signaling pathway.
