[objective]To study the pathological changes of renal injury induced by fluorosis in rats with different kinds of calcium preparations,and to select the best calcium preparations to relieve fluorosis.[methods]Fifty male SD rats(SPF rats)were randomly divided into five groups according to their body weight: Control Group(basic diet,distilled water),fluoride-attacking group(basic diet,100 mg/L Na F water)and calcium carbonate-attacking group(1% Ca CO3,100 mg/L Na F water).Serum Calcium,phosphorus,creatinine and urea nitrogen were measured by colorimetry after 90 days of feeding in the groups treated with fluoride-supplemented calcium hydrophosphate(1%Ca HPO4,100 mg/L Na F water)and calcium gluconate(1% calcium gluconate(CG),100 mg/L Na F water).The expression of Bax,Bcl-2,ROCK I and Cyto-C protein in the kidney of rats was observed by HE staining.The relative m RNA expressions of Bax,Bcl-2,ROCK I,Cyto-C,Caspase-6,Caspase-7 and Apaf-1 were analyzed by RT-PCR.[Results] the results showed that compared with the control group,the kidney tissue in the fluoride attack group showed obvious vacuolar degeneration,the interstitial small vessels dilated,the expression of Bax,ROCK I and Cyto-C protein increased significantly(p<0.05),and the expression of Bcl-2 protein decreased significantly.The relative expression of Bax,ROCK I,Cyto-C,Caspase-6,Caspase-7 and Apaf-1 genes increased significantly,while the relative expression of Bcl-2 gene decreased significantly.[Conclusion] The experiment shows that fluoride can damage the kidney of rats.Supplement of calcium can alleviate the kidney damage of fluorosis to some extent,and the effect of Calcium gluconate is better. |
