Noise has been a common occupational and environmental hazard.Hearing loss due to overexposure to noise is the second most common types of sensorineural hearing loss.Previous studies demonstrated that antioxidant reagents have been shown to protect the cochlea from noise-induced damage.Hydroxysafflor yellow A(HSYA)is the major active chemical component of the flower of the safflower plant named Carthamus tinctorius L.Previously,it has shown to possess various effects,especially neuroprotection against cerebral ischemia-reperfusion(I/R)injury was reported by anti-oxidant action and anti-apoptotic action.In this study,we aimed to evaluating effects of HSYA on noise-induced cochlear injury and possible signaling pathways in mice.Our results showed that HSYA significantly alleviated hearing loss and suppressed heir cell death in noise-exposure mice.We found that HSYA decreased the expression of cox-2,which is related to oxidative reaction.Using RNA-sequencing,it was revealed that the expression of caspase-3 in the control group is much higher than HSYA group.Additionally,HSYA inhibited the expression of protein Cox-2 and caspase-3.This study provides that HSYA protect NIHL,to certain extent,by enhancing the antioxidant defense system and exerting anti-apoptotic effects.
