ObjectiveTo investigate the relationship between single nucleotide polymorphism of SMG6 gene rs216172 and genetic susceptibility to coronary heart disease in the Koreanpopulation in Yanbian area, and to provide basis for the study of genetic mechanism of coronary heart disease in Korean population in this area.MethodsIn this paper,a case-control study was used to 244 patients with coronary heart disease in Yanbian Korean population diagnosed in the cardiovascular department of Yanbian University between march 2017 and november 2018,with an average age of 64.27±9.56 years,of which 138 were male and 106 cases were female.A total of 233 healthy subjects of the same age and sex who underwent physical examination at the physical examination center during the same period were randomly collected as controls, the average age of 64.45±9.01 years old, male 125 cases, women 108 cases.Subjects' history of hypertension, diabetes mellitus, smoking,BMI,blood lipids and blood glucose were collected. The typing test of SNP adopted SNaPshot technology, and the biochemical indicators were detected on the roche automatic biochemical analyzer.Statistical analysis of data using Excel 2007 and SPSS 23.0.The measurement data are described by the mean ± standard deviation, and the comparison of the mean numbers adopts the Independent sample t test. The counting data is expressed as a percentage or rate, and the comparison of the rate adopts the Chi-Square test.Result(1)Comparison of general data of research objectsThe history of hypertension, smoking history and TG in coronary heart disease group were significantly higher than that in control group(P=0.022,P=0.015,P=0.023),while HDL was significantly lower than that in control group(P=0.032),and there was no significant difference between the two groups in age, sex,BMI,systolic pressure, diastolic pressure, history of diabetes mellitus,TC,etc.(P>0.05).(2)SNP Analysis of rs216172 sites in coronary heart disease Group and control group? The frequency of C and G alleles in coronary heart disease Group of Korean population in Yanbian area was 66.8%and 33.2%respectively,the frequency of C and G gene in control group was 73%and 27%respectively,and the frequency of G allele in coronary heart disease group was significantly higher than that in control group(P=0.038);Compared with the control group, the frequency of 3 genotypes(CC?GC,GG)in rs216172 loci was significantly different(P=0.011),and the CC genotype of coronary heart disease was lower than that of the control group.?When the GG+GC genotype of the rs216172 loci of the SMG6 gene was dominant model, the coronary heart disease group was significantly higher than that of the control group, and the difference was statistically significant(P=0.004).?There was no significant correlation between genotype frequency and allele frequency of rs216172 loci and hypertension and diabetes mellitus(P>0.05).(3)Logistic regression analysis showed that GG+GC genotype at rs216172,hypertension,smoking were independent risk factors for coronary heart disease(P<0.05).Conclusion1.The rs216172 loci of SMG6 gene are related to the pathogenesis of coronary heart disease in Yanbian Korean, and its G allele is a risk factor for the pathogenesis of coronary heart disease.2.GG+GC genotype at rs216172?hypertension and smoking are independent risk factors for coronary heart disease in Korean population in Yanbian area.
