Rhodiola sachalinensis is a perennial herb or subshrub plant.In China,it is not only a traditional Chinese medicine,but also applied to functional foods in recent years.It has obvious therapeutic potential for diabetes that currently harms peopled physical and mental health.The microorganisms in food were killed by appropriate dose of irradiation treatment and storage of food was facilitated,and at the same time the composition and content of the compounds were changed.In this paper,the effects of different doses of 60Co-γ ray irradiation on the composition and content of active constituents of ethanol extracts from Rhodiola sachalinensis(REE)and its hypoglycemic effect were studied,which provided a new reference for the development and utilization of Rhodiola sachalinensis resources.It provides new ideas for the application of irradiated Rhodiola sachalinensis in hypoglycemic.The Rhodiola sachalinensis used in this experiment was irradiated at the Sichuan Institute of Atomic Energy,with 60Co-γ rays as the radiation source,and the irradiation doses were set to 5,10,20 and 30 kGy,respectively.The composition and the content of the main compound of REE before irradiation(irradiation dose of 0 kGy)and after were analyzed by high performance liquid chromatography(HPLC),followed by animal test and a-glucosidase inhibition assay to select irradiation dose which better hypoglycemic effect,and finally use REE which by this irradiation dose treatment to study drug dose on the hypoglycemic effect by animal test.In the animal experiment,streptozotocin(STZ)was used to induce C57BL/6 male mice to establish type I diabetes model.After successful modeling,mice were gavage with the REE which irradiated with different irradiation doses,and the body weight,fasting blood glucose level,food intake and water intake of the mice was measured regularly.And determination of oral glucose tolerance on the last day,rapid dissection after eyeball blood withdrawal,weighing the heart,liven spleen and kidney,using kit to determine plasma total cholesterol(TC),total Triglyceride(TG),glycated hemoglobin(GHb),malondialdehyde(MDA),catalase(CAT),and superoxide dismutase(SOD).The a-glucosidase inhibition assay was performed using a 96-well plate method with acarbose as a positive control.Compared with the non-irradiated(irradiation dose was 0 kGy),the content of salidroside in the REE was significantly decreased after different doses of 60Co-γ-ray irradiation(P<0.05).No tyrosol was detected when the irradiation dose was 5,10 and 30 kGy.The content of rosavin was significantly increased when the irradiation dose was 5,10and 20 kGy(P<0.05).When the irradiation dose was 20 kGy,tyrosol was detected and detected a compound different from other irradiation doses.The REE treated with different doses of 60Co-γ-rays all recovered the weight gain of mice,which reduced the blood sugar level,food intake and water intake of mice and significantly improved the food utilization rate(P<0.05),significantly reduced TC,TG,GHb and MDA levels(P<0.05)and significantly increased CAT activity and SOD activity(P<0.05),and reduced the phenomenon of organ swelling in mice,and the effect was better at 20 kGy,and there was a significant difference(P<0.05)with the other irradiation doses.In addition,the 20 kGy 60Co-y-ray irradiated REE for the inhibitory effect of the activity of a-glucosidase was significantly better than the other irradiation doses(P<0.05).The 20 kGy 60Co-y-ray irradiated REE had a dose-effect relationship with the hypoglycemic effect of type I diabetic mice,and it was better when the drug dose at 280 mg/kg.Compared with non-irradiation,the composition and content of compounds in REE after different doses of 60Co-γ-ray irradiation were changed.The REE before and after irradiation could improve the basic metabolic and blood parameters of type I diabetic mice,relieve heart,liver and kidney swelling,and inhibit α-glucosidase activity,the effect was better when the irradiation dose was 20 kGy.And the hypoglycemic effect had a certain dose-effect relationship,and the high dose was better.
