Study On The Regulatory Effect Of Chinese Gallnut On Immune Function Of CD4~+T Cells In Transgenic K14-VEGF Psoriasis Mice Based On The Hoop Circumference

objective:By studying the effect of Chinese gallnut ointment on the content of IL-4 and IFN-?in peripheral serum and the change of IL-17 in skin lesions of transgenic K14-VEGF psoriasis mice to discuss the regulatory effect of gallnut ointment on immune function of CD4+T cells,and elucidate the mechanism of gallnut in the treatment of psoriasis.Methods:Selecting 60K14-VEGF psoriasis mice randomly,and they were randomly divided into 6groups(blank group,vaseline group,tazarotene group,50%gallnut ointment group,25%gallnut ointment group,10%gallnut ointment group).Then 10FVB mice were randomly selected as a group.After grouping,the ear lesions were labeled,and the corresponding cream was smeared to the ear lesions of mice through the hoop circumference in twice a day for 4 weeks.The blank group and the FVB group were not treated.After 4 weeks,using ELISA to test the levels of IL-4 and IFN-?in serum,and using immunohistochemical method to test the level of IL-17 in lesions were detected.The statistical software SPSS20.0 was used to describe and infer statistics,and quantitative data were expressed by mean and standard deviation.Comparisons between groups were tested by One-Way analysis.The intra-group comparison was tested by LSD test.The test level?was 0.05,the difference is considered to be statistically significant when P<0.05.Results:1.After treating for 4 weeks,we studied that gallnut ointment can effectively ameliorate the ear lesions and histopathological features of transgenic K14-VEGF psoriasis mice,of which 50%gallnut ointment has the best effect;2.The serum levels of INF-?in gallnut ointment groups and tazarotene group were lower than those in blank group(P<0.05),and the serum levels in 50%gallnut ointment group and tazarotene group were significantly lower than those in control group(P>0.05);3.The gallnut ointment groups and tazarotene group has no effectively influence of the serum levels of IL-4,and there was no difference among the groups(P>0.05);4.The treatment groups can reduce the level of IL-17 in psoriasis mice(P<0.05),but there was no improvement in Vaseline group(P>0.05),there was also no significant difference between 50%gallnut ointment group and tazarotene group(P>0.05).Conclusion:1.we found that gallnut ointment can effectively improve the ear lesions and histopathological changes of transgenic K14-VEGF psoriasis mice;2.The gallnut ointment can decrease the level of INF-?in peripheral serum of model mice,50%gallnut group had the most obvious effect,but the effect on IL-4 was not obvious;3.The gallnut ointment can effectively reduce the level of IL-17 in skin lesions of psoriasis mice;4.The therapeutic effect of gallnut cream on psoriasis model mice maybe realized by adjusting the imbalance of Th1/Th2 and Th17/Treg to regulate the immune function of CD4~+T cells.
