CircRNA is a kind of highly conserved double-stranded closed ring non-coding RNA.Different from common non-coding RNA such as microRNAs,circRNA plays an important role in organisms.It can specifically inhibit the function of microRNAs by binding to microRNAs,thus affecting the biological behavior of cells,thereby affecting the upstream and downstream signaling pathways.In the early stage of the laboratory,10 microRNAs(let-7e,mi-21,mi-34 a,mi-122,mi-146 a,mi-148 a,mi-148 b,mi-152,mi-130 a and mi-130b)that may interact with Wnt1 gene were found by collating the reported data and bioinformatics prediction methods.Seven circRNA targeting these microRNAs were predicted by bioinformatics methods,and these circRNAs were compared with two.There are four signaling pathways related to TGFbeta: circRNA_BTBD7,circRNA_MRPS35,circRNA_PHIP,circRNA_RFC1 and three targeting FAK-related signaling pathways: circRNA_EXOSC10,circRNA_SLC25A3 and circRNA_PSMD1.This topic focuses on the seven circRNA related functions.OBJECTIVE: To investigate the effects of seven circRNA such as circRNA_BTBD7 on the biological functions of cancer cells by targeting and binding to microRNAs.METHODS: Seven target fragments of circRNA mature region such as circRNA_BTBD7 were ligated with pCD2.1 cyclic overexpression plasmid to construct the overexpression vector and transfected into SaoS-2 and MG63 cells to observe its overexpression before and after transfection.RESULTS:(1)Cell biology experiments showed that a series of circRNA had significant effects on cell proliferation,migration,invasion and apoptosis.Among them,circRNA_PHIP could inhibit osteosarcoma cell proliferation and regulate cell cycle,circRNA_EXOSC10 could promote cell apoptosis,circRNA_PSMD1 could promote cell proliferation,circRNA_SLC could promote cell proliferation.25A3 can inhibit apoptosis.(2)Bioinformatics analysis and double luciferase reporter gene experiments confirm that circRNA binds to different microRNAs.MicroRNA-148a/microNA-148 b,microNA-152/microNA-3666/microNA-4295 and other microRNAs are the aggregate targets of a series of circRNA.CONCLUSION: CircRNA_PHIP can inhibit the proliferation of osteosarcoma cells and regulate cell cycle,circRNA_EXOSC10 can promote cell apoptosis,circRNA_PSMD1 can promote cell proliferation,and circRNA_SLC25A3 can inhibit cell apoptosis.These functions may be achieved by combining multiple microRNAs with Wnt and TGF-beta signaling pathways.
