Objective and Significance:As one of the most common malignant tumors in the world,gastric cancer is a serious threat to human life and health.China is a high-risk area for gastric cancer.In the cause of death of malignant tumors in China,gastric cancer ranks second.At present,the comprehensive treatments of gastric cancer mainly include surgical resection,radiotherapy,chemotherapy,targeted therapy,and immunotherapy.However,the prognosis is not very optimistic,and the 5-year overall survival(OS)rate is less than 30%.Invasion and metastasis of tumor cells directly affect the treatment and prognosis of gastric cancer.Tumor metastasis is a complex and multi-step process regulated by multiple links and multiple factors.Infiltration and metastasis of tumor cells to surrounding normal tissues is achieved through the basic pathway of extracellular matrix(ECM)degradation.Related studies have shown that matrix metalloproteinases(MMPs)play an important role in this procession.MMP-2 and MMP-9,which are important members of the MMPs family,have been most closely related to the invasion and metastasis of gastric cancer.Traditional Chinese medicine has remarkable clinical effects in the treatment of tumors,especially in stabilizing tumors,reducing tumor recurrence,reducing the adverse effects of radiotherapy and chemotherapy,improving the quality of life(QOL)of patients,and enhancing the immunity of patients.Xiao'ai Jiedu Prescription was created by the great TCM master professor Zhou Zhong-ying,based on his long-term clinical practice and experience,and under the guidance of the"cancerous toxin"academic theory.Xiao'ai Jiedu Prescription has been proved to have significant advantages in cancer treatment.Previous experimental studies have confirmed that Xiao'ai Jiedu Prescription has remarkable anti-cancer effects on lung cancer and liver cancer,but its effect on gastric cancer remains to be further investigated.As an indispensable herbal medicine of this anti-cancer prescription,ophiopogonin B has various pharmacological functions including anti-tumor effects.Steroidal saponins,high isoflavones,polysaccharides,and amino acids are the main compounds isolated from Ophiopogon japonicus.Among them,ophiopogonin B(OP-B)is a steroidal saponin monomer component,which has been reported to show obvious anti-cancer effects.The purpose of this experiment was to evaluate the effects of OP-B on proliferation,invasion,migration,and apoptosis of human gastric cancer cell MGC-803,and to determine the possible molecular mechanism.Methods:The poorly differentiated human gastric cancer MGC-803 cells were selected as our research objects by consulting relevant literature.Human gastric cancer MGC-803 cells were cultured in vitro,and the effect of OP-B on its proliferation was observed by CCK-8 method.The invasion and migration ability of human gastric cancer MGC-803 cells was observed by invasion assay and wound-healing assay,respectively.Western Blot technique was used to verify the effect of OP-B on the protein expression of MMP-2 and MMP-9 in human gastric cancer MGC-803 cells.Annexin V-FITC/PI double staining combined with flow cytometry was used to detect the effect of OP-B on the apoptosis rate of MGC-803 cells.Results:1.The results of CCK-8 assay showed that OP-B significantly inhibited the growth of human gastric cancer MGC-803 cells in a dose-dependent manner,after treatment of 24 h and 48 h.2.After the treatment of OP-B on human gastric cancer cells MGC-803 for 24 h,invasion ability of cells was significantly inhibited in a dose-dependent manner,compared with the control group.It was observed under the microscope that the number of invaded cells in the treatment groups was significantly less than control groups.3.The results of wound-healing assay showed that OP-B significantly inhibited the migration ability of MGC-803 cells in a dose-dependent manner.4.Western Blot assay demonstrated that OP-B could down-regulate the expression of MMP-2 and MMP-9 in MGC-803 cells.5.OP-B significantly enhanced apoptosis of MGC-803 cells.Conclusions:1.OP-B significantly inhibited the proliferation of human gastric cancer MGC-803 cells.2.OP-B could inhibit the invasion and migration ability of MGC-803 cells.3.The anti-tumor effects of OP-B in part via down-regulation of the expression of MMP-2 and MMP-9 proteins in MGC-803 cells.4.OP-B significantly increased apoptosis rate in MGC-803 cells,showing that the anti-cancer effect of OP-B may also be associated with other apoptosis-related pathways.
