Comparison of biosafety of five kinds of dentin adhesivesObjective:To compare the biosafety of five clinically used commercial dentin adhesives,and to explore the influencing factors of the differences and their relationship with the degree of conversion of monomers.Methods:1.The cytotoxicity of five commercial dentin adhesives was measured by MTT method.The five kinds of dentin adhesives were cured,and then the extract was immersed.The immersin g time of the extract was 1 day,3 days and 5 days.The human dental pulp cells were extracted by tissue block enzymatic digestion,and the cells of the fifth generation during logarithmic growth phase were used to detect the cytotoxicity of the dentin adhesives.Morphological evaluation of human dental pulp cells was performed by fluorescent staining techniques.The morphology of human dental pulp cells was observed and photographed by the fluorescence inverted microscope.Human dental pulp cells cultured in five different kinds of dentin adhesive extracts were compared with normal human dental pulp cells.Comparison between groups was also performed2.Degree of conversion of five kinds of dentin adhesives was measured by FT-IR.The degree of conversion values of five kinds of dentin adhesives were compared in pairs,and compared with the cytotoxicity results to investigate the relationship between the biosafety and the degree of conversion of five kinds of dentin adhesives.Results:1.Five kinds of dentin adhesives exhibit different cytotoxicity.With the prolonged immersion time of the extract,the cytotoxicity of the adhesives also changed accordingly.Prime&Bond,Clearfil SE Bond,OptiBond FL all showed no cytotoxicity,and Single Bond 2 group showed mild cytotoxicity.For the Adper Easy One group,there was no cytotoxicity at 1d,mild cytotoxicity at 3 days,and moderate cytotoxicity at 5 days.Morphological evaluation of human dental pulp cells showed that the morphology of living cells in Adper Easy One group changed generally and the other four groups had good morphology when compared with the control group.2.The degree of conversion values of five kinds of dentin adhesives were different,ranging from about 35%to 81%.Among them,Prime&Bond had the highest degree of conversion value,while Clearfil SE Bond had the lowest detection value.Conclusions:1.Different kinds of dentin adhesives have different biosafety.2.The biosafety of different kinds of dentin adhesives has a certain correlation with the degree of conversion of monomers,and there are many influencing factors.
