Mechansim Discussion Of JiDeSheng Sheyao Inhibiting Hepatocellular Carcinoma Through Regulating Hsa_circ₀013290

Background:Hepatocellular carcinoma?HCC?is one of the most frequent malignancies.It is the fifth mostcommon cancer and the third cause of cancer-related death worldwide.Hepatocellular carcinoma rates are the highest in East and South-East Asia,with China alone accounting for about 50%of the totalnumber of cases and deaths,and the incidence is on the rise,which is a serious threat to human health.Early detection,early diagnosis and early treatment are the key to improve the curative effect of HCC.In the recent years,increasing evidences identify circular RNAs?circRNAs?as a class of important regulators in various human cancers.Nevertheless,the functions and mechanisms of most circRNAs remain largely unknown.We will select hsa_circ₀013290 to explain its possible role in of HCC,and to evaluate its value in the diagnosis,treatment and prognosis of HCC.Ji DeSheng Sheyao is a traditional Chinese medicine,which has the functions of clearing heat and detoxifying,activating blood circulation and removing stasis,swelling and relieving pain.Previously,it is mainly used to treat snake wound.Recent clinical studies have shown that it also plays a certain role in the treatment of hepatocellular carcinoma.We proposed to explore the role of Ji DeSheng Sheyao in hepatocellular carcinoma from the circRNA level,in order to provide new ideas for the diagnosis and treatment of hepatocellular carcinoma.Objective:To explore the value of hsa_circ₀013290 in the diagnosis and treatment of hepatocellular carcinoma and the effect of Ji DeSheng Sheyaomedicated serum on HCC.Methods:qRT-PCR was used to detect the relative expression of hsa_circ₀013290 in 50 HCC patients.The relationship of the expression with HCC patients as well as clinical pathological parameters were futher analyzed.qRT-PCR was used to detect the regulation function ofJi DeSheng Sheyao medicated serum on the relative expression of hsa_circ₀013290 in SMMC-7721 cells.The effect of Ji DeSheng Sheyao medicated serum on SMMC-7721 cells proliferation was tested by CCK-8 and EdU assays.The effects of Ji DeSheng Sheyao medicated serum on SMMC-7721 cellsapoptosis and cell cyclewere tested by flow cytometry.Results:In 50 patients with HCC,the relative expression of hsa_circ₀013290 in tumoral tissues was significantly lower than that in adjacent tissues?P<0.01?.The relative expression of hsa_circ₀013290in tissues were positively correlated with the tumor size and clinical stages?P<0.05?,and were not correlated with gender,age,AFP,HBV and Pathological stages?P>0.05?.The relative expression of hsa_circ₀013290 in SMMC-7721 cells of 20%Ji DeSheng Sheyaomedicated serum group was significantly higher than that of the control group?P<0.01?.Compared with the control group,the cell proliferation of the experimental group was significantly inhibited?P<0.01?,and the apoptosis rate increased?P<0.01?,the effects were positively correlated with the Ji DeSheng Sheyaomedicated serum concentration.However,Ji DeSheng Sheyao medicated serum showed no effect on the cell cycle of SMMC-7721?P>0.05?.Conclusions:The relative expression of hsa_circ₀013290 in tumoral tissues was significantly lower than that in adjacent tissues,suggesting that hsa_circ₀013290 plays a tumor suppressor role in the development of HCC.Ji DeSheng Sheyao can inhibit the proliferation and induce the apoptosis of SMMC-7721 cells and regulate the relative expression of hsa_circ₀013290 in SMMC-7721 cells,providing a new idea for the treatment of HCC.
