Objective: By establishing the model of rat endometriosis,we observed the effect of Chinese medicine Neiyitongjingling decoction on the expression of TGF-?1,T?R?,Smad2/3 and Smad7 in the heterotopic model of endometriosis rats,Further explore the pathogenesis of EMs and provide a theoretical basis for Neiyitongjingling decoction treatment of EMs.Methods: Healthy female non-pregnant Spragne-Dawley(SD)rats(body weight 190-210 g)were selected as subjects,and were randomly divided into 6 groups,7 in each group,which were blank control group(normal sham operation group)and model control group.the high-dose group of Neiyitongjingling,the middle dose group of Neiyitongjingling,the low dose group of Neiyitongjingling and the western medicine group(gestrinone capsule).The blank group of rats underwent open abdominal sham surgery,the other groups of rats were replicated into a model of endometriosis,and the model was tested for the fourth week.After the fourth week,Rats in blank control group and model control group were given saline by intragastric administration.The rats in the high,middle and low dose groups of Neiyitongjingling were given different concentrations of Chinese herbal medicine by intragastric administration.Rats in western medicine group were given a certain concentration of gestrinone solution by intragastric administration.Four weeks later,rats in eutopic(blank control group only)and ectopic endometrium tissue were extracted and killed.HE staining,immunohistochemistry and Western blot were used to detect the expression of TGF-beta 1,T?R?,Smad2/3 and Smad7 in eutopic(blank control group)and ectopic endometrium tissues of rats.The protein expression between the groups was compared.Results: 1.After 4 weeks of treatment,the volume of ectopic endometrium in the high,middle and low dose groups and the western medicine group was significantly lower than that in the model control group,P<0.05,the difference was statistically significant;There was no significant difference in ectopic endometrial volume between the various dose groups and the western medicine group,P>0.05.2.The endometrial tissues of rats in the model control group were observed to grow vigorously by HE staining.The ectopic endometrial glandular epithelial cells of the different doses of Yitongtongjingling and western medicine group showed different degrees of atrophy.3.Compared with the model control group,the TGF-?1,T?R?,Smad2/3 protein content in the high and medium dose Chinese medicine groups and the western medicine group were significantly decreased,and the Smad7 protein content was significantly increased.P<0.05,the difference was statistically significant.The protein expression levels of the high and medium-dose Chinese medicine groups and the western medicine group were compared,P>0.05,and the difference was not statistically significant.Conclusions: 1.Neiyitongjingling decoction can significantly reduce the volume of ectopic endometrium and inhibit the growth of endometrium,so as to achieve the purpose of treating endometriosis.2.Neiyitongjingling decoction can reduce the protein expression of TGF-?1,T?R? and smad2/3,and increase the protein expression of Smad7,so as to achieve the purpose of treating endometriosis.3.Treatment of endometriosis can be achieved by modulating the expression of TGF-?/Smads signaling pathway-related factors.
